Piglet’s E. coli is an important infectious pathogen which harm piglets, over the years, due to not normative use of the veterinary clinical drug resistance in production were generalized. In recent years, studies have shown that the drug resistance of bacteria have a lot to do with bacterial biofilm, bacterial biofilm can be affected by some of the Traditional Chinese medicine ingredient, which mediates antibiotics into the bacterial biofilm, kill bacteria inside the biofilm, improve the cure rate of clinical infectious diseases. It is highly resistant to current piglet E. coli has its positive clinical significance.This study intends to e. coli biofilm model was established using the orthogonal experiment, and then use five experimental drugs for e. coli biofilm inhibition experiment, finally to piglets with yellow-white dysentery as experimental animals to observe the curative effect of different drugs group.E coli was cultivated after experiment choose clinical bacteria separation and identification,then E coli in different concentrations、different cultivation time and different temperature were used as factors that influence the E coli biofilm formation by orthogonal test table L9(34), enzyme-labeled instrument measuring the OD value as indicators for E coli biofilm (BF)to find the best forming conditions.Physiological and biochemical experiments results show that the separation of bacteria of E coli was successful, scanning electron microscopy (sem) shows that the E coli BF was more successful under the optimal conditions of culture, the optimal conditions for E coli OD mean value was0.450, RSD was0.76%.Experiments using the double dilution method to determine the MIC of E coli, after E coli BF was formed successful, use different drug groups:I group for florfenicol, II group for Fructus Schisandrae Chinensis, III group for ShuangHuang-Lian, Ⅳ group for florfenicol unite Fructus Schisandrae Chinensis,V group for florfenicol unite ShuangHuangLian to compare each group to observe the effect of E coli BF. Test results show that florfenicol has minimal MIC value of E coli, Fructus Schisandrae Chinensis was the largest. And relative to the drug use alone, V group had best inhibition effect to E coli BF (p<0.05), and reached73.82%±0.15%, the effect to E coli BF of Ⅳ group was stronger than used alone for florfenicol and Fructus Schisandrae Chinensis,had reach66.63%±0.18%.Experiment select breeding pig200head which has yellow-white dysentery on lactation piglet farm, randomly divided into5groups, each group with40, then testing the effect of different drugs treatment group comparison, drugs were grouped according to the determination of E coli SMIC method. Result shown that V group piglet mortality rate was only5%, lower than other experimental group, while florfenicol group rates were highest. Piglet V group cure rate had reached92.5%, far higher than other group.Above results show that the separation of E coli bacteria was successful identified and proved, which provides an experimental platform for subsequent experiments. Through the orthogonal experiment to screen the optimum culture conditions of E coli BF made the the experimental plan simple and feasible greatly, and saves time and cost; Florfenicol as excellent antibacterial chemicals, MIC to E coli was least was match the expected results, but in the observation of different drugs for E coli BF inhibition effects suggest that traditional Chinese medicine combined with chemical drugs was better than when used alone Chinese medicine or chemical drugs. Florfenicol combined with ShuangHuangLian application of injection in the treatment of piglets with yellow-white dysentery have obvious effect, shows that the method has great clinical applied potential in yellow-white dysentery of piglets disease.
