Traditional feed has rich in resistant digestion adhesive mannan and xylan, due to thelivestock and poultry of digestive enzymes of excluding mannase, animals themselves hasdifficult to completely degrade xylan, Therefore using such substances into transformationand absorb need exogenous enzymes, but there is another added problem with enzymedirectly of high cost.With adding enzyme producing probiotics approach is simple, can beachieved to improve feed utilization and reduce farming costs.The study of screening by Congo red plate changing colour circle method to producexylanase and mannanase production of Bacillus subtilis, by liquid fermentation and assayof enzyme activity screening, and animal gastrointestinal tolerability and safety testing,screening out safety subtilis strain can tolerate the animal gastrointestinal environment andhas high enzyme activity. Strain B-5producing xylanase and strain NB-5producingmannanase are the final selection. The B-5strain and NB-5strain in pH2still maintainedbetter, the survival rate reached90%, are also well tolerated in artificial intestinal fluidsimulation environment.By morphological characteristics, physiological and biochemical properties and16srDNA sequence analysis, strain B-5and strain NB-5are identified as bacillus subtilis, aresafe are safe the Ministry of agriculture feeding animals strains. The test of coexistencebetween two strains, confirm the existence of not antagonistic strains, can be made intocompound inoculants at the same time feeding.The production process of bacterium agent is studied. With strains of spore formationrate and spore number for the determination of index, respectively on two strains of singlefactor and orthogonal experiment, spore production best condition of the two strains isobtained.B-5strains: corn flour2%, yeast powder3%, MgSO4·7H2O0.3%, K2HPO40.01%,KH2PO40.02%, speed200RPM, Loading100/250mL, quantity of6%, the fermentationtime of96h,NB-5strains: corn flour1%, yeast powder1%, CaCl20.1%, K2HPO40.01%, KH2PO40.02%, speed200RPM, Loading100/250mL, quantity of4%, the fermentation time48h。 Under the condition of the nutrition and training, the preparation of the two trains of acteriaagent, has paved the way for the production of bacteria strains agent.The valley ducks as test animal, B-5strain, NB-5strain and inoculum agent twocomposite agent feeding application effect was investigated. Through the changes ofconventional nutrients were measured and the growth performance, the additive group ofducks, the change of intestinal flora of intestinal digestive enzyme activity change and inthe stool, inquiry agent to promote ducks growth performance and mechanism ofimproving feed conversion ratio. The results show that: adding the growth performance ofinoculum on the duck had significant promoting effect, daily body weight of ducks than inthe control group were improved in different degree, an average increase from4.58%to7.85%, feed to gain ratio were decreased in different degree, an average increase from3.10%to7.41%; agent can be on trial for the composition of intestinal bacteria duck groupwere adjusted, the test group of intestinal beneficial bacteria Bifidobacterium andLactobacillus are different the number increased, diarrhea caused by E.coli and otherharmful bacteria decreased; In cellulose, enzyme activity and mannase vitality xylanase ofexperimental group improved than blank group, other enzymes, protease, lipase andamylase, etc.) than the blank group were increased; common nutritional substances such ascrude fiber,crude protein, crude fat and starch than in the control group were also lower inexperimental group in feces. That enzyme agent can improve the transformation andutilization of feed ducks. The result of this study provides a new way for efficient breedingduck, provides a solid theoretical foundation for improving the technological level of duckbreeding.
