| [objective]Secretory immunoglobulin A, SIgA is one of the IgA which was firstly found by Chodircker and Tomasill] in1960’s. Because of the difference of sourcedistribution,structure,function; IgA is formed two systems, one is Secretory immunoglobulin A, the other is serology immunoglobulin A. The combination and secretion of SIgA is in the intestines, respiratory tract,lacteal gland and salivary gland, so SIgA which mainly exist in the secretion of gastrointestinal tract and bronchus plays a part roll on the anti-infection. SIgA is essential in protecting mucosal by ensuring immune exclusion. It plays an important roll on the body’s immunization.This experiment observes the the influence of hexadecadrol, IL-21, estradiol benzoate on the SIgA content and the areas of SIgA positive plasma cells in the intestines of mouse vaccinated by salmonella O antigen.[Methords]Female health BALB/c mice aged6weeks were randomly divided into4groups with16mice in each. physiological saline group, hexadecadrol group, estradiol benzoate group,inteinterleukin-21grou. Each mouse was fed Salmonella O antigen. Each mouse in estradiol benzoate group was injected estradiol benzoate liquid. Each mouse in hexadecadrol group was injected hexadecadrol liquid.Each mouse in IL-21group was injected IL-21liquid. Each mouse in physiological saline group was injected physiological saline. After7days,do all the steps once again.After14days each mouse was sacrificed. We will put duodenum of these mice in the4%density paraformaldehyde stationary liquid,and then immobilization, entrapment,slice cut. The duodenum were tested for the areas of SIgA+plasmocyte by immunohistochemical analyze. We’ll cut0.5g intestinal mucosal tissure on the icy plate. And the it will be put in the physiological saline to homogenate,which is six weight more than the intestinal mucosal tissure. After3000r/min centrifugate for20minutes and then abstract supernate. It will be stored in the-20℃temperature.The levels of SIgA in the intestinal of the mice were determined by ELISA.[Results]In estradiol benzoate group, the conte of SIgA is13.53±0.867ng/ml.In physiological saline group, the contet of SIgA is7.233±0.616ng/ml. The difference is so large (P<0.05)In IL-21group, the contet of SIgA is13.83±0.4ng/ml. In physiological saline group, the contet of SIgA is7.233±0.616ng/ml.
