Rumex crispus is the perennial herb of the genus Polygonaceae in the family Rumex L.It is also named Dock root etc. Its main function is clearing away heat and toxic materials,relaxing the bowels, mule killing etc. There is a tiptop curative effect on treat metrorrhagiapsoriasis. Recently people scoop it a lot because of its important medicinal value. And withthe environmental degradation this herb becomes less and less. This research begins withcallus induction growth differentiation and differentiation of bud rooting in order to protectthis kind of wild-lived herb. And finally the asexual regeneration system of Rumex crispuswas established successfully. In the same time it has been detected emodin in the wild plantsRumex crispus.The main content of this research and research results were as follows:1、The differentiation of growing pointThe research showed that the most suitable way to differentiate the growing point was todifferentiate after rooting in the liquid medium. And the optimum medium for different ofgrowing point was MS+6-BA2.0mg·L-1+NAA0.05mg·L-1+sucrose30g·L-1.2、The induction, multiplication and differentiation of Rumex crispus callusWhen there was a research on the callus induction we found that the best material forcallus induction was petiole, the optimum medium was MS+6-BA2.0mg·L-1+2,4-D0.2mg·L-1+NAA0.2mg·L-1+sucrose30g·L-1. The optimum medium for callus multiplicationwas MS+6-BA2.0mg·L-1+2,4-D0.4mg·L-1+sucrose30g·L-1, for callus differentiation wasMS+ZT0.6mg·L-1+NAA0.4mg·L-1+AgNO30.6mg·L-1+sucrose30g·L-1.3、The cultivation of budinitiation rootingThe result of bud rooting showed that the optimum medium for rooting was MS+IAA0.2mg·L-1+sucrose20g·L-1and the most suitable condition was constant temperature andillumination. The percentage of rooting was100%and the rooting number was6.35averagethe length was10.54cm average.4、Transplanting and colonization of test-tube plantlet There was no process of refine seedling when the sprout had been transplanted. The ratesof survival were100%in both indoor and outside. The survival plantlet grew exuberant andretained all biological characteristics of the escape Rumex crispus.5、The analysis of emodinThe result of analysis on the root plantlet and callus showed that all the parts of plantletcontains emodin. The content was respectively7.70%,3.08%and6.24%in root plantlet andcallus.
