Chitinase (EC3.2.1.14) is a kind of enzyme to catalytic chitin hydrolysis reaction togenerate N-acetylglucosamine. Many plants, including herbaceous and woody plants canproduce chitinase. In recent years, anti-fungal disease has become a research focus, andchitinase plays an important role in the resistance of plants to fungal diseases, researchon chitinase has gained more and more popularity by the majority of scientists. With thein-depth research, it turns out that chitinase enzyme not only act vital role in plantdevelopment, withstand adversity and symbiotic nitrogen fixation, but also is related toanti-fungal diseases of plants. as a result the determination of the activity of chitinase andfiltering out high chitinase activity wheat is of great significance to enhance China’swheat production.Taking231wheat varieties as test material, we test chitinase activity byspectrophotometry at420nm, anaysis wheat chitinase activity at4℃, and select thewheat, the main conclusions are as follows:1.Wheat varieties chitinase activity were significantly different. Afterlow-temperature treatment, chitinase activity increased in varying degrees, and theincrease was significant difference between varieties.2.Before and after low temperature treatment with higher chitinase activity werewheat varieties: CP20-3-1-4-2, Wesfarmers889, Chuanmai42(red), Chuannong43, thenew wheat89019, Chuannong21, Ning0076, CP99-13-6-3-1-1, new original958,51180,Conon181, total11varieties, these resources can be used as the resistant parent.3.After low temperature treatment chitinase activity increased considerably varieties:Huaiyin0454, Stone02-6207,876, Ning0453, Wesfarmers889,02P67, MY13B16, newwheat89019, Ning0078, Yang01-116, stone03-Y119, total11varieties, indicating thatthese germplasm can make better reflect to low temperature stress, so there should be arelatively high breeding value.4.Higher chitinase activity before low-temperature processing after treatmentincreased considerably higher varieties Xinong889,02P67,ning0078,the new wheat89019, these four species are very resistant germplasm, has greater breeding value.
