Jujube belongs to Rhamnaceae Zizyphus. Jujube is one of important economic treespecies and dried fruit,also is one of the most ancient and unique species in our country,there is8000years of cultivation history. Because it is easier to manage, earlier to fruit,nutritious and versatile, there is more than30countries introduce it and spread it to all overthe world. In China, many areas and provinces have planted Jujube tree exceptHeilongjiang province. In Shanxi, Hebei, Shandong,Henan, Shaanxi province, jujubecultivation area is very large, it has already become an important part of agriculturalproduction,and played a positive role to the prosperity of the rural economy andimprovement of people’s living standards.Xishan JiaoZao is Anhui Chizhou traditional specialty fruit tree species,and Jujubecultivation was harmed by Jujube witches broom. Early in1984, professor Wang Guyuanof Anhui agricultural university horticulture college proposed to use paliurus hemsleyanusas stock to resist JWB. over the years,it has been confirmed that paliurus hemsleyanus asstock has the ability to resist JWB. Today the methed is still widespread used in the jujubecultivation because it can effectively prevent the JWB, and make the trees grow moreprosperous, fruit quality improve.New shoots of paliurus hemsleyanus stock and Xishan JiaoZao stock were used as testmaterials. Combined with characteristics of the plant material itself, new shoots samples ofpaliurus hemsleyanus vaccination stock with control group and jujube stock vaccinationgroup with control group were respectively prepared by using metabonomics technologythrough metabonomics analysis of Jujube tree new shoots’ metabolites. To get processedaqueous phase after grinding, weighing, extraction, derivatization in jujube new tips.According to NIST and Wiley Database, metabolic flow changes of these metabolites inthe process of jujube growth and development were detected by using GC–MS for sampleanalysis, then the main differences of metabolites by different treatmen were compared.These research work will provide basis to research and reveal theJWB disease resistancemechanism.Established the metabolic spectrum analysis technology platform of the aqueousextract of Chinese jujube new tip, and analyzed the selected samples in water phase ofChinese jujube new tip through GC–MS. It turned out that the discrimination of thechromatographic peak and the separation effect of Chinese jujube new shoots in each processing degree is very good, and many kinds of saccharic acid has been effectivelyseparated, confirming the derivative method is proper and feasible, and the optimizedchromatographic conditions are suitable for Chinese jujube new tip samples in water phase.Retrieving and identifying the metabolites in the graph one by one by using the NIST massspectral database, I identified183peak from the separated mass spectrum peak, then Icame to a conclusion that the main ingredients of aqueous extract of Chinese jujube newtip are sugars, glycoside, alcohols, organic acids and amino acids.Through PCA and PLS analysis of the GC-MS detection data of the aqueousextract of Chinese jujube new tip in each processing degree, it shows that each dealedsamples can be distinguished, and this indicates that the difference of the constitution ofmetabolic substance in each processing degree isn’t obvious. However, the distributionform of the same kind of block is almost similar, it makes clear that material compositionof the paliurus hemsleyanus block and Chinese jujube is very different. The intensity ofsamples of Chinese jujube new tip at different development stages which are in sameprocessing is good, it shows that the repeatability of this experiment is perfect.Based on PLS-DA analysis of the extract of Chinese jujube new tip, I obtainedthe VIP component information after verification, the first20metabolites componentsarranged according to the size of the VIP value mainly includes sugars, glycoside,alcohols, organic acids and amino acids. The results indicate that the sugar acidmetabolism and the mino acid metabolism of Chinese jujube changed dramatically afterthe new tip samples of the paliurus hemsleyanus block Xishan JiaoZao and the blockXishan JiaoZao were grafted Jujube witches broom, it provides an important basis forrevealing which metabolic pathways of Chinese jujube change after the Jujube witchesbroom invading, and this has very important sense in the research of the disease-resistantmechanism of Jujube witches broom in jujube.
