In recent years, poultry industry develop rapidly, but immune inhibitory diseasepopular seriously, make the disease more challenged and difficult to control, tostrengthen the dependence on vaccines and antibiotics, at the same time, reduce ofthe immune effect on vaccine and abuse of antibiotic make the drug resistance ofpathogen stronger and other shortcomings are gradually exposed, new green immuneenhancer and immune adjuvant arises at the historic moment. Polysaccharide asmaterial sources, low cost, efficacy significantly, and to solve drug residues andcontrol disease has obvious advantages, be the first choice of the new immuneenhancer. Polysaccharide is composed of multiple monosaccharide condensation,water loss of macromolecular substances, are highly complex structures and speciesspecificity and modern research proves that polysaccharides have immunomodulatory,antitumor, anti adhesion, resistance to oxidation, the effect of lowering blood sugar.Sofar, many plant polysaccharide has been shown to have immune enhancement effect.Phoenix tree flower, as the Verbenaceae Glorybower flower, according to"supplement to compendium of Materia Medica" records, the flower has the effectof dispelling the wind, depressurization,curing durchfall with high medicinal value. Atpresent, our country’sPhoenix tree flowers’ planting gradually formed the scale, themedicinal value of the flower has attracted wide attention, and many results have beenachieved, but the domestic research of the Phoenix tree flower only on thecomposition of the extraction and analysis, human health products research anddevelopment, but research on Phoenix tree flowers’polysaccharide used in preventionand control of livestock and poultry disease is rarely reported. The naturalenvironment and geographical features of Taishan provide a convenient to theplanting in large scale.Therefore, the research group Taishan Phoenix tree flower asthe research object, collected fresh pine pollen in the spring from Taishan area, sievedbroken water extraction and used alcohol precipitation method for extraction of crudepolysaccharide. In the extraction process,used pepsin and Sevage reagent twomethods of removing protein to guarantee the high purity of crude polysaccharide,selected by the SPF embryo hatching chicks as animal model of health to discussPhoenix tree flower polysaccharides’ function of enhancing immune of chicks. Weaimed at the development of new plant immune enhancer and making contribution forthe development of health animal husbandry. This research consists of two parts as follows:Part One: Effects of Taishan Firmiana simplex flowers polysaccharide onproliferation of splenic lymphocytes in chickenTo explore the Taishan Firmiana simplex flowers polysaccharide (TFSFPS), SPFchicken spleen lymphocytes in vitro and the test can be divided into two parts: thefirst part is the two groups of chicks that were injected with three differentconcentrations (400μg/mL200μg/mL100μg/mL) TFSFPS and setting Taishan pinepollen control group (TPPPS a concentration of400μg/mL) and blank control group.Preparing spleen lymphocytes at the first day,14days,21days, chicken spleenlymphocytes was measured by PI stainingconversion rate; In the second part,the thethree concentrations TFSFPS were added to the chicken splenic lymphocytes thatwere isolated and cultured in vitro, setting the same TPPPS control group, after48h,chicken spleen lymphocyte transformation rate was measured by PI staining. Theresults showed that, in the splenic lymphocyte proliferation assay in vivo, the role ofTPPPS and three concentrations TFSFPS spleen lymphocyte transformation rate wassignificantly higher than the blank control group, but less than TPPPS control group;In the spleen lymphocyte proliferation test, the two polysaccharides group spleenlymphocyte transformation rate was also significantly higher than the TPPPSgroup,lymphocyte transformation rate is higher than the concentration TFSFPS group.Thus, TFSFPS chicken splenic lymphocytes have a stimulating effect in and outsidevivo of the proliferation, but the stimulation is weaker than TPPPS. The test resultsprovides a theoretical basis that TFSFPS can be developed into a novel immuneenhancer.Part Two: Immune enhancement on chicken of Taishan Firmiana simplexflowers polysaccharideTo study the effects of immune enhancement caused by the Taishan pine pollenpolysaccharides on chick and the effects of immune regulation on immunosuppressedchick, we selected two hundred and twenty-five healthy chick that hatched by SPFembryos and they were randomly divided into9groups: Ⅰ,Ⅱ,Ⅲwere the TFSFPSgroup,injected of TFSFPS400mg/mL,200mg/mL,100mg/mL by the necksubcutaneous injection respectively; Ⅳ,Ⅴ, Ⅵ were the TFSFPS and CTX group,injected of three concentration polysaccharide and40mg/kg CTX meanwhile; Ⅶwas the CTX control group, only injected of40mg/kg CTX; Ⅷ was the TPPPS control group, injected of400mg/mL TPPPS;Ⅸ was the control group, injected ofequal sterile saline.All the chicks were vaccinated with ND vaccine and the relevant indexes such asthe immune organs index, the serum antibody levels,the amount of leukocyte,lymphocyte ratios, lymphocyte transformation rate,intestinal SIgA levels, IL-2andIFN-γ were detected after vaccinated3d,7d,14d,21d,28d,35d,42d. The results showedthat all the immune indexes of group Ⅰ、Ⅱ、Ⅲ、Ⅷwere significantly higher thangroup Ⅳ、Ⅴ、Ⅵ、Ⅶ、Ⅸ(P<0.05),indicating that TPPPS had the effects of immuneenhancement on chick and relieved the immunosuppressive to some extent. Amongthe four groups Ⅰ、Ⅱ、Ⅲ and Ⅷ, Ⅷ group were higher than the other threegroups, but without significant difference compared with group (P <0.05).
