Infectious diseases of livestock and birds are the chief problem harm to the development of livestock farming, especially viral diseases; however, there was no effective medicine to cure viral infectious diseases and people pay more attention to the drug residue and side effects of chemical medicine. Recent years, with the development of science and technology people's living standard being continuous improved. More and more people concern the product'safety relation to health and environment, such as food, medicine, cosmetic, etc.; they want more green food and livestock product. The natural traditional Chinese medicine (TCM) displays definite advantages: comes from nature, has low or no side effect, no residue, coming into more people's vision and more scientists are researching and developing new natural medicine from TCM instead of chemical medicine in preventing and curing viral livestock and birds diseases.Many researches have showed that a lot of traditional Chinese herbal medicinal ingredients have antiviral activity and immunologic enhancement. In this stuty we studied Lonicera japonica Thunb. and Flos lonicerae which have a long history as a good heat-clearing and detoxicating medicine in China. We extracted volatile oil by Supercritical CO2 fluid extraction, chlorogenic acids and flavonoids by different concentration ethanol combining ultrasonic wave from the three TCMs. The extracts'antiviral and immune-enhancing action and mechanism were investigated through a series of experiments in vitro. The purpose is to compare the pharmacological action of Lonicera japonica Thunb. and Flos lonicerae and offer theory basis for developing herbs as anti-virus praeparatum.We get volatile oil 1.71g, 4.17g, 1.55g from two types Lonicera japonica Thunb. and one Flos lonicerae witch habit separately is Fengqiu County Henan Province, Longhui County Hunan province, and Xupu County Hunan province. The extraction rate separately is 1.22%, 2.78%, 1.03%. The three Chinese herbals'chlorogenic acids content separately is 2.72%, 4.06%, 3.14%, and flavonoids content separately is 5.55%, 4.33%, 5.17%.Extracts of the three Chinese herbal medicines within safety concentration added into Vero cells monolayer before, after and simultaneous with PRV and NDV respectively. Cytopathic effect (CPE) and MTT absorption method are employed to illustrate anti-virus effect. The cells OD values were determined in order to evaluate the effects of herbs on virus infecting cells. The results showed that the three different extracts: volatile oil, chlorogenic acids and flavonoids of three herbs inhibit infectivity of the PRV, NDV to Vero cells significantly, depending on their concentrations. They could decrease CPE lesions, inhibit virus to infect the cell and neutralize virus in dosages dependent; they behaved in inhibiting virus directly or promoting cell antivirus or both. It suggested that earlier administration should have better efficacy as clinical application. There are no significant different antiviral effects between Lonicera japonica Thunb. and Flos lonicerae. It can be used in clinical veterinary drugs in place of expensive Lonicera japonica Thunb. for cheep Chinese herbal Flos lonicerae.Effects of the extracts on proliferation of chicken splenic lymphocytes were investigated by the methods of MTT colorimetric assay. The results showed that the extract could promote proliferation of chicken splenic lymphocytes significantly (p<0.05), which were induced by ConA or LPS, and the effects were related to concentration of the extracts.
