Vibrio anguillarum is the causative agent of a highly fatal haemorrhagic septicaemia known as vibriosis, which can cause great economic loss in aquaculture industry. Therefore, the detection of V. anguillarum is particularly important in the prevention and control of vibriosis. The current reports on detection of V. anguillarum can not be applied in fish farms and other grass-roots units because of the disadvantage of time-consuming and complex detection operation. In this study, antibody labeling technology and rapid immunochromatographic assay were used to prepare a test strip for V. anguillarum. The colloidal gold, quantum dots and phosphorescent material were used to label antibody of V. anguillarum.The V. anguillarum MVM425was used in monoclonal antibody perparetion, and the monoclonal antibodies (MAbs)5G4-G4and5G4-B8were obtained. The colloidal gold was used to label MAb5G4-G4and prepare the rapid test strip for V. anguillarum. The test strip had a good specificity and no cross-reaction to the common pathogens. The detection limit of the strip was105CFU/ml.It was reported that the detection sensitivity could be greatly improved by using quantum dots and up-conversion phosphor as antibody markers. In this study, the availability of core-shell quantum dots CdSe/ZnS and up-conversion phosphor NaYF4:Yb, Er as antibody markers in the detection of V. anguillarum was investigated. By optimizing the combination of antibody and these two materials, quantum dots and up-conversion phosphor material labeled monoclonal antibody were obtained. Then, the feasibility of its application in the detection of V. anguillarum was verified by dot-blot assay. The results indicated that quantum dots and up-conversion phosphorescent material could be used in V. anguillarum detection, and this study provided useful information for the development of quantum dots and up-conversion phosphor test strip for V. anguillarum.
