Paramisgurnus dabryanus is a Small-scale economic fish in our country, and it has great potential of export. In recent years, with the increase of the Fishing intensity. The wild resources is reducing Gradually. Aquaculture boom of Paramisgurnus dabryanus is set of in our country. But now the seed of the Paramisgurnus dabryanus is come from the Wild-caught,the excellent seed is scare. Therefore,in order to develop the Aquaculture of Paramisgurnus dabryanus in a Sustained, stable and healthy environment,it’s Imperative to start the breeding work of the Paramisgurnus dabryanus. We Intends to adopt Cross-species amplification and Database mining to develop the SSR of the Paramisgurnus dabryanus. And we will collect some geographical populations Paramisgurnus dabryanus to measure some morphological parameters to anlysis the differences。And we will anlysis the Genetic Diversity of the different populations with the SSR.Then we do the diallel cross in thedifdferent populations and get the F1generation,we can measure the growth traits of the F1generation to assess the effct. The study aims to lay a foundation for future selective breeding of the Paramisgurnus dabryanus. The main findings of this study are as follows:1. Dabryanus microsatellite markersFrom published articles microsatellite primer sequences obtained in47loach, were screened out of the10pairs with type clear polymorphic microsatellite primers better for dabryanus groups cross-species amplification, and each pair of PCR primers to amplify the annealing temperature was optimized primer name, sequence and annealing temperature. Agarose and detected.10microsatellite loci amplified dabryanus groups are good, PCR products were PAGE electrophoresis can produce clear bands. The results showed that the number of alleles10microsatellite loci ranged, on average contain9.9alleles per locus from8-12; effective number of alleles were less than the number of alleles, between4.1388between9.5410. Observed heterozygosity range of10microsatellite loci was0.0256~0.5500, with an average of0.3459; while the expected heterozygosity in the range of0.7682~0.9074, with an average of0.8577. In this study, the preliminary view that filter out10pairs of microsatellite markers Loach is applicable dabryanus can serve as a good marker of its genetics research.2. Analysis of diversity. Genetic different geographic populations of dabryanusIn dabryanus four geographic populations were detected were detected95alleles per locus detected an average of9.5alleles. The average number of alleles in a population of5.200to10.000, polymorphic information content sites PIC at0.546~0.803, is moderately to highly polymorphic loci, alleles least Henan groups, the largest group of Anhui expected heterozygosity HE groups in0.767~0.874, the lowest group in Henan, Anhui highest population, with an average of0.836. Groups observed heterozygosity (Ho)0.242~0.327in the lowest group Hubei, Hunan highest population, with an average of0.284. A number of alleles and expected heterozygosity HE Anhui groups are the largest. Thus, the level of genetic diversity in populations of higher Anhui groups in other regions. Fixed index Fis showed four groups showed heterozygous deletion (Fis>0). Hardy-Weinberg equilibrium test showed that all groups have at least five loci deviated from Hardy-Weinberg equilibrium (P<0.05). In Mac229-SSR, Mac425-SSR and Mac477-SSR loci all groups have deviated from Hardy-Weinberg equilibrium (P<0.05). According to DA genetic distance construct phylogenetic trees, UPGMA clustering shows four geographical groups mainly clustered into three categories, Hubei, Anhui, genetic distance smaller first clustered together, after the Hunan group together as a class, and then a separate group in Henan clustered together.3. Reproduction and offspring growth traits with the force of different geographic populations of dabryanusIn seven different hybrids or crosses from the Hubei population groups selfing progeny obtained full-length, body length, body height, body thickness and weight maximum, significantly higher than the other combinations. Followed by the progeny population groups within Henan selfing obtained. As population growth trait value Henan and Liaoning female population as a male hybrid progeny obtained full-length, such as a minimum. Analysis of variance showed that paternal, maternal and paternal maternal mean square have reached a significant level (P<0.01), described the parents of the full-length, body length, body height, body GCA thickness and weight (existed between GCA) effects a very significant difference, and specific combining ability (SCA) there are very significant genetic differences. GCA is the overall average of the average yield of a parent and the other parent dubbed several F1F1and all combinations of the difference between SCA analysis refers generally with a particular combination of the average of their parents force difference. The former is mainly affected by additive effect, the latter dominant and epistatic effects of genes affected. The results showed that male full-length, body length, body height, body thickness GCA and weight (GCA) effects of population values in the order of Hubei, Hunan, population, population in Henan, Anhui populations, Liaoning populations. Female full-length, body length, body height, body thickness and weight GCA values were from large to small populations in Hubei, Henan populations. Hubei male population GCA values are mostly positive, indicating that the use of dabryanus Hubei easier with a male as with a growth advantage combinations. In the specific combining ability analysis, the relative effect of Henan population crosses maximum value, followed by the Hubei population crosses the smallest population in Henan and Hubei as female as male population combination.
