| The purpose of this study was to investigate the effects of cold stress on rats under simulatedtransport stress, and hope To study the effect of cold stress resistance added VC, VE and Gln inthe diets of rats. A cold-stress model being established on Wistar rats.110clean female wistar rats were randomly divided into three groups by L9(34), theexperimental group was intragastric administration mixture of VC, VE and Gln, other twogroups were divided into cold stress group and normal temperature group. normal temperaturegroup were raised under temperature of21±1℃. Cold stimulation groups were exposed to thetemperature of11±1℃for24h after intragastric administration mixture of VC, VE and Gln. Theconcentration of antioxidant parameters in serum (NE, GSH-PX, SOD, T-AOC, MDA) weredetected by ELISA technology. The concentration of immune indexes in serum (IL-2, IgA, IgG,IgM) were also detected by ELISA technology. The relative expression of HSP70in lymphocytewere screened by qRT-PCR and western blotting technology. Then further data analysis byprincipal component analysis (PCA) and consolidated balance method. The results were showedas following:(1) Effect of Vitamin C, Vitamin E and Glutamine compound nutrients on serum NE andbiochemical parmeters.The concentration of NE in serum generally tended to increase after24h of cold stress. TheNE concentration test results showed that the experimental group was extremely significantlydifference compared with the cold stress group (P<0.01). Group6NE level with normaltemperature group showed no significant change. The concentration of SOD in serum generallytended to reduce after24h of cold stress. The experimental group has highly significantdifference with the cold stress group (P<0.01).2,3with8,4with7,5with6groups had nosignificant difference. The concentration of GSH-PX in serum generally tended to reduce after24h of cold stress(P<0.01). The experimental group has rising compared with the cold stressgroup.8,9groups compared with normal temperature group had no significantdifference(P>0.05), Other groups showed significant difference compared with normaltemperature group (P<0.01). The concentration of T-AOC in serum generally tended to reduceafter24h of cold stress(P<0.01), the experimental group has rising compared with the cold stress group.1,2,3,4,5,8,9groups compared with each other had no significant difference, buthas highly significant difference with the cold stress group (P<0.01). The concentration of MDAin serum generally tended to increase after24h of cold stress (P<0.01), the experimental grouphas reduced compared with the cold stress group.5group compared with normal temperaturegroup had no significant difference, others compared with cold stress group has highlysignificant difference(P<0.01).(2) Effect of Vitamin C, Vitamin E and Glutamine compound nutrients on immune indexes inserum.The concentration of IL-2in serum generally tended to reduce after24h of coldstress(P<0.01). The results of immunoglobulin(IgA, IgG, IgM) in serum:The concentration of immunoglobulin in serum generally tended to reduce after24h of coldstress(P<0.01). The experimental group has rising compared with the cold stress group. Group6IgA level with group7showed no significant change, others compared with each other hassignificant difference(P<0.05). Group8IgG level with normal temperature group showed nosignificant change, and6with7groups had no significant difference,3with5groups hadsignificant difference(P<0.05). Group4,6IgM level with cold stress group showed nosignificant change (P>0.05),1,7,8groups showed significant difference with cold stressgroup(P<0.05), and3,5groups showed no difference with normal temperature group (P<0.05).(3) Effects of VC, VE and Gln compound nutrients on HSP70expression of lymphocyte ofrats under cold stress.The HSP70mRNA level of q-PCR test results showed that the cold stress group wasextremely significantly difference compared with the normal temperature group, generallytended to increase after24h of cold stress (P<0.01), and compared with the other groups showedincreased extremely significantly (P<0.01).6,8,9groups showed significant difference withnormal temperature group (P<0.05), others showed highly significant difference with normaltemperature group(p<0.01), but has reducing in common. The HSP70expression changesshowed as follows, the cold stress group increased extremely significantly compared with normaltemperature group (P<0.01),8,9groups showed significant difference with normal temperature group (P<0.05), others showed highly significant difference with normal temperaturegroup(p<0.01), but has reducing in common. These results were the same as HSP70mRNAlevel.(4) Selection of optimal formula for VC, VE and Gln compound nutrients.The raw data of the11indicators were analysis by principal component analysis (PCA) andconsolidated balance method. The results showed that the best intragastric administration dosewere VC:300mg/kg; VE:100mg/kg; Gln:2500mg/kg.The above results proved that the VC, VE and Gln compound nutrients has a good effects oncold stress.
