Steviol glycoside is a new natural sweetener and it is been extracte from the leaves of stevia rebaudiana which belongs to Composite plants. It has unique properties of high sweetness and low calorie. The sweetness of it is300times than cane sugar, but only1/300calorie. It is known as "the third source of sugar in the world". The objective of the present study is to develop a method for rapid determination of the content of stevioside (St) and rebaudioside A (RA) in Stevia Rebaudiana leaves. In this study, the effect of radiation mutagenesis of stevia by heavy ions have been researched. Then screening the cold resisant mutants by the radiation of heavy ions. The major results were as follows:1. The study of the effects of Stevia by heavy ions:The Demonstration shows the feasibility of heavy ion irradiation of Stevia Breeding by detection of stevia seed germination rate, survival rate of seedlings, seedling height and seedling shoots and other indicators of physiological activity. Which laid the foundation for the next step of screening cold.2. The screening of the cold resistant mutants by the radiation of heavy ions:The identification of C80-1and C80-3has better cold tolerance by the growth rate of seedlings and physiological activity.3. The study showed that SLS can be used to extracted spectra information thoroughly to analyze the contents of St, the correlation coefficients of calibration (rc) and prediction (rp), the root-mean-square errors of calibration (RMSEC) and prediction (RMSEP), and the residual predictive deviation (RPD) are0.959,3.48,4.39and3.87. These of the optimal model for RA are0.924,3.99,4.23and3.6.
