Study On X\Y Sperm Separation And Sexed Semen Production Of Dalian Black Cattle

It is a more effective method to control animal sex using sexed semen for artificial insemination and sexed embryos production, while the most widely used way to produce separated sperm is improved flow cytometry. Here we tried to establish technical process for sexed sperm freezing of Dalian black cattle by adjusting sperm separation technology parameters, which is based on the sexed semen production process of Holstein and the characteristic of Dalian black cattle semen.Results:The purity of sexed Dalian black cattle semen could reach to more than89%(89%～91%) using flow cytometry at a certain separation speed; the sperm motility of Dalian Black cattle sexed semen after thawing was significantly improved by adjusting glycerol concentration in diluents; the sperm motility and survival time of thawed Dalian black cattle sexed semen after thawing was improved when adding antioxidant VE and CAT in freezing dilution; the conception rate was58.3%(235/403), and male calves proportion was93.2%(219/235)using sexed semen for artificial insemination.Conclusion:This research reported that we established the sexed semen production and the technical process of Dalian black cattle sperm separation by adjusting sperm separation technology parameters and semen freezing method, based on Holstein sexed semen production technology, and also carried out the demonstration application of small scale industries. Our study showed that it was feasible to produce sexed semen of Dalian black cattle using MOFLO.
