| In the experiment, we established an A FABP transgenic mice model to determineA FABP gene expression level and its impacts on fat depositions of different organizationsand other functional genomics.First, a cow A FABP gene overexpression vector was built and transfered into mices. Theoverexpression vector was obtained from pEGFP c1, which was digested with HindIII/BamHI and inseted with optimized cow A FABP gene. After linearized, the treated vector wastransfered into mice by microinjection technology. Thus, A FABP transgenic mice had beenbuilt. To verify the functional properties of A FABP gene, parental mice and their offspring F1,F2, F3generation were tested with growth and development indicators, serum biochemicalparameters, Fourier transform infrared spectroscopy, transgenic mice tissue sections, gene andprotein levels of A FABP.The results were showed as following:1. A FABP gene was expressed stably in mice;2.A FABP gene didn’t affect the normal growth, development and reproduction of mice.3.A FABP gene promoted the deposition of fat in mouse tissue and increased levels of body fat.4. A FABP gene was transfered stably between various generations of mice.In conclusion, A FABP gene can increase fat deposition in mice and can be expressed invivo stability without causing other functional genomics changes. Establishment of A FABPtransgenic mouse model will provide a scientific basis for the establishment of transgenicanimals in the future. |
