Objective:1. To explore the concentration of Glycogen, Lactic Acid LD, inorganic phosphate Pi and ATP in gastrocnemius on daurian ground squrriels and14-day-hindlimb rats study the effects of disuse on skeletal muscle Fatigue related biochemical indexes.2. To test TP50, TPT, RT50, RT75, P0and Pt study the effects of disuse on skeletal muscle contractile properties;3. To explore the the expression amount of SERCA1and M-CK mRNA study the effects of disuse on the function skeletal muscle SR;4. To test fatigue in gastrocnemius on daurian ground squrriels and14-day-hindlimb rats study the effects of disuse on skeletal muscle fatigue resistance.5. To study the effects of Tetramethylpyrazine on14-day-hindlimb rats skeletal muscle fatigue related biochemical indexes.Methods:1. The concentration of Glycogen, Lactic Acid LD, inorganic phosphate Pi and ATP determined by ultraviolet spectrophotometer;2. The skeletal muscle contractile properties tested by PowerLab/400;3. SERCA1mRNA and M-CKm RNA was tested by Real Time PCR.Results:1. Skeletal muscle fatigue related biochemical indexes of ground squrriels Compared with interbout arousal group the Glycogen of pre-hibernation, hibernation and post-hibernation increased55%,56%and57%(P<0.01), respectively; Lactic Acid LD has no significant change (P>0.05); Compared with pre-hibernation, inorganic phosphate Pi in hibernation decreased35%(P<0.05); Compared with pre-hibernation ATP in hibernation, interbout arousal and post-hibernation decreased58%,65%and62%(P<O.01) respectively;2. Skeletal muscle contractile properties of ground squrrielsCompared with pre-hibernation,in interbout arousal and post-hibernation RT50increased by37%and43%(P<0.05), RT75increased by48%(P<0.05) and62%(P<0.01);3. Skeletal muscle fatigue of ground squrrielsHibernation has no significant effects on the fatigue of ground squirrel gastrocnemius;4. Expression amount of SERCA1and M-CK on ground squrrielsCompared with pre-hibernation, the expression amount of SERCA1in Hibernation, interbout arousal and post-hibernation decreased by50%,41%and45%(P<0.05); M-CK increased119%(P<0.05) in interbout arousal;5. Skeletal muscle fatigue related biochemical indexes of ratsAfter14-day-hindlimb unloading,the Glycogen, Lactic Acid LD of gastrocnemius in rats were increased49%and37%(P<0.01), inorganic phosphate and ATP decreased24%(P<0.05) and58%(P<0.01),respectively;compared with hindlimb unloaded group the hindlimb unloading plus intragastric Ligustrazine instillation Lactic Acid LD of gastrocnemius decreased23%(P<0.01).6. Skeletal muscle contractile properties of ratsAfter14-day-hindlimb unloading, Pt and P0decreased29%(P<0.01) and17%(P<0.05) respectively, RT50and RT75decreased35%and34%(P<0.05) respectively.7. Skeletal muscle fatigue of ratsAfter14-day-hindlimb unloading increased fatigue in rats gastrocnemius;8Expression amount of SERCA1and M-CK on rats After14-day-hindlimb unloading, the expression amount of SERCA1mRNA in gastrocnemius increased49%(P<0.05) and SERCA2mRNA decreased60%(P<0.05)Conclusion:1. Glycogen in interbout arousal has a significantly decrease, Lacticv Acid LD has no significant change in any phases, inorganic phosphate Pi is lower in hibernation and ATP is higher in pre-hibernation; After14-day-hindlimb unloading, Glycogen, Lacticv Acid LD increased significantly, norganic phosphate Pi and ATP decreased significantly;2. Hibernation significantly increased RT50and RT75;14-day-hindlimb unloading Po and Pt significantly decreased, RT50and RT75also significantly increased;3. The different period of hibernation the fatigue of gastrocnemius has only less change.14-day-hindlimb unloading the fatigue of gastrocnemius significantly increased;4. During different period of hibernation SERCA1mRNA relative expression showed a significant decrease.14-day-hindlimb unloading, SERCA1mRNA relative expression showed a significantly increase;5. Plus intragastric Ligustrazine instillation can inhbit the increasing of LD.
