Neospora caninum is an intracellular protozoan parasite that causes themovement dysfunction of dogs, bovine abortion and other symptoms. It has a widehost range which promote the spread of infection Neospora caninum, and broughthuge economic losses in livestock farming. There haven’t proved the Neosporacaninum can infect humans, but studies have shown that Neospora caninumantibodies are detected in human serum.And the Neospora caninum can be cultured ina variety of human cells in vitro, so it can’t be ignored. There is on effective drugs andvaccine to treatment Neosporosis,and the effective way of preventing and controlNeosporosis is testing the infected cattle, then eliminated, to the establish a noNeospora caninum infected cattle. Accurate diagnosis is an effective method ofcontrol the spread of the disease, so the research in diagnosis and epidemiologicalinvestigation of Neospora caninum is very important. This experiment has establishedthe nested PCR detection method,and detected dogs, pigeons in changchun area. Thisaritle mainly includes the following sections:1. Neospora caninum NcP40specific gene (NO. NCLIV034390) was used into design specific nested PCR primers and establish a nested PCR method, and allreaction conditions are optimized with annealing temperature at56℃and Mg2+concentration at1.5mmol/L.This conditions has high specificity and sensitivity thatcan detect a parasite gene, and does not cross-react with Toxoplasma gondii.Hammondia hammond DNA.2. An epidemiological investigation of Neospora caninum was carried out ondogs and pigeons in Changchun by nested PCR. The infection rate of210pigeonbrain tissue samples was30%, ITS1sequence analysis results show that there are5genotypes,40of them in agreement with the NC-PR,10agree with NC-1, the restof the three are new genes for has not been reported. The infection rate of120dogfeces samples was41.7%, and the Neospora caninum infected in dogs has fourgenotypes,28agree with NC-PR,13agree with NC-1, the rest two are newgenotype. New genes are evolved by the NC-PR. The experiment established a Neospora caninum NcP40-specific nested PCR anddetected Neospora caninum in pigeons and dogs in Changchun. The establishednested-PCR can be used to detect Neospora caninum and epidemiologicalinvestigation, we also get some Neosporosis epidemiological data.
