Objective:To observe renal interstitial pathological changes and the expressionof transforming growth factor beta1(TGF-β1) and α-smooth muscle actin(α-SMA) inrenal tissue of unilateral ureteral obstruction(UUO)mice by therapeutic interventionwith Huangkui capsules,and to investigate the effect of Huangkui capsules on renaltubulointerstitial fibrosis.Methods:Km male mice54, the average body weight is18-22g. The mice wererandomly divided into sham operation group (n=18),UUO group (n=18) andHuangkui capsules treatment group (n=18).They were scarificed on3,7,14days afterestablishing UUO model.Obstructed kidney were examined by HE staining,Massonstaining, immunohistochemisty staining and reverse tanascription-polymerise chainreaction(RT-PCR). Renal interstitial pathological changes and the expression ofTGF-β1and α-SMA in renal tissue were observedResults:(1) Our results showed that the renal interstitial damage and renalinterstitial fibrosis area were significantly increased in UUO group and treatmentgroup compared with the sham operation group (P<0.05), and reduced in treatmentgroup compared with UUO group(P<0.05).(2) Expression of the TGF-β1and α-SMAwere signify-cantly increased in UUO group and treatment group compared with thesham operation group (P<0.05), and increased in UUO group compared withtreatment group (P<0.05).(3) The expressive levels of TGF-β1mRNA and α-SMAmRNA were significantly increased in UUO group compared with the othertwo groups (P<0.05). The expressive levels were significantly increased in treatmentgroup compared with the sham operation group (P<0.05).(4) Correlate analyzeshowed that TGF-β1and α-SMA were positively correlated with renal interstitialpathological changes(P<0.05).Conclusion:TGF-β1,α-SMA and renal tubulointerstitial fibrosis are closelyrelated; Huangkui capsules can significantly reduce the expression of TGF-β1andα-SMA,thus it could suppress fibrosis of renal interstitial. |
