ObjectiveUsing immunohistochemistry and TUNEL method and image analysis to observe theexpression of NF-kBp65and TERT in normal endometrium, eutopic and ectopicendometrium with Adenomyosis. To discuss the the role of NF-kBp65and TERT and theirrelationship in the development of adenomyosis.Methods1. Selected the endometrium、lesions tissues with adenomyosis as eutopic group、ectopicgroup; the endometrium with Uterine fibroids、CINⅢ as controls.2. Detect the NF-kBp65in the three groups, Then using Leica Q500MC image analysissystem to dectect the intensity of NF-kBp65expression.3. Detect the TERT in the three groups,Then count the number of apoptotic cells underthe Optical microscope.4. Using SPSS13.0statistical software to analyze the results.Results1. NF-kBp65was mainly expressed in the cytomembrane of normal、eutopic and ectopicendometrial gland cells. Their PU were121.60±21.21、139.00±5.96、166.76±16.55, the differencewas statistical significance among the three groups(P <0.05).2. The PU of secretory phase(123.90±4.37134.64±6.54)was higher than proliferativephase (119.64±5.31、131.71±7.67) in the controls and eutopic group, the difference wasstatistical significancel in the controls(P <0.05), but made no sense in eutopic group. Theproliferative phase、secretory phase of controls and eutopic group compared with statistical significance(P <0.05).3. TERT was mainly expressed in the nucleus of normal、eutopic and ectopic endometrialgland cells. Their AI were3.40±5.51、13.36±6.34、15.44±4.67.The difference between controlsand eutopic group、controls and ectopic were statistical significance (P <0.05);but had nostatistical significance between eutopic group and ectopic group (P>0.05). The AI ofproliferative phase(2.84±1.86)、secretory phase(1.52±0.43)in the controls compared withstatistical significanc(eP <0.05); And the AI of proliferative phas(e13.27±5.09)、secretory phase(12.78±5.92) in the eutopic group compared with no statistical significance(P>0.05).Theproliferative phase、secretory phase of controls and eutopic group compared with statisticalsignificance(P <0.05).4. In the development of adenomyosis, There was a correlation between NF-kBp65andTERT(γ=0.435,P=0.03).Conclusion1.NF-kBp65、TERT expressed in eutopic and ectopic endometrium with Adenomyosis werehigher than normal endometria. It prompt that the both were involved in the development ofadenomyosis.2. In eutopic group, NF-κBp65and TERT all had high expression in proliferative phase、secretory phase, show that in the two stage endometrial cells have a stronger ability to invadethe myometrium and cause the adenomyosis.3.In the development of adenomyosis,There was a negative correlation between NF-kBp65and TERT.
