Bone marrow mesenchymal stem cells (BMSCs) are the promising pluripotent stemcells in the clinical application, which originated in the embryonic mesoderm. previousstudies demonstrated that BMSCs almost can be obtained in each individual organism,and can differentiate to GABAergic neurons with conditioned induction. At present, stemcells are often mentioned: embryonic stem cells, neural stem cells, umbilical cord bloodstem cells and bone marrow mesenchymal stem cells. For clinical application,transplantation of autologous stem cell is the safest way, and the best seed stem cellsshould have the outstanding ability of differentiated potential, and has a reliable source.Due to the ethical constraints, the embryonic stem cells is not the proper choose, thesource of neural stem cells is scarce, and umbilical cord blood stem cells can not be save.These kinds of cells above are not appropriate as a source of autologous transplantation ofstem cells. And bone marrow mesenchymal stem cells not only have potential ofdifferentiation, the source materials is more stable and convenient, so we choose bonemarrow mesenchymal stem cells as the seed cells, the separation and purification of bonemarrow mesenchymal stem is relatively simple from adult and child. BMSCs is the most ideal autologous stem cells.Epilepsy has been a common and frequently-occurring disease inneuroscience,According to the statistics,There are millions of epilepsy patients inChina.Epilepsy has become the second most common diseases in neurology,its morbidityis just below the incidence of headaches.In the process of the morbid development,the lostof GABAergic neurons is the main pathological change.Currently,the main clinicaltreatments of epilepsy are drugs,surgery and neural control,etc.Most are symptomatictreatments for Abnormal discharge.Although these methods can control seizures invarying degrees,radically treat epilepsy is need the re-newing of CNS.An important question is how to increase the differentiation rate of the GABAergicneurons from BMSCs, So,we want to explore the differentiation mechanism,We havetaken note of Mash1,which is important in regulating the nervous system development.Inthis study,We choose Mash1as the target genes of the experiment.We reduce theexpression of Mash1by RNAi.Then induce the differentiation of these cells, we found thatthe differentiation rate reduced.
