Objective To test the effect of transplanting bone stromal stem cells modified by green fluorescent protein to the hippocampus of epileptic rats,.then observeing and inquirying to the survival and immigrating of the cells and the integration with and repair the brain tissue,and to test the effect of the cells of the apoptosis in the epileptic rats.To provide some theoretical bases on the therapy of epilepsy with cells transplantation.Method The epileptic activity was produced by penicillin injection .Rats were divided into A: normal control group(n=8) ,B: group of the epileptic without treatment (n=8) , C: group of the epileptic with Sodium Chloride (n=24) ,D: group of the epileptic with bone marrow stromal stem cell (n=24) .①the epileptic model is build by penicillin peritoneal injection.②Bone marrow stromal stem cells of transgenic mice with green fluorescent protein were isolated and cultured.③The 5ul of cells or sodium chloride were respectively grafted into the right hippocampus of the rats with the stereotaxic apparatus.Then the rats were respectively perfused, drawed the materials, cut sheet, staining at 7 days, 14 days,28 days after grafting.④The survival and immigration of bone marrow stromal stem cells were observed in the fluorescence microscope.⑤The morphous of neuron apoptosis is observed by HE staining in the epileptic brain.⑥The number of the apoptosis cells and the expressing of the bcl-2 protein and bax protein were detected by immunohistochemical stain. The neuron apoptosis is known by compare the BMSCs group with sodium chloride group,⑦The ambulatory electroencephalogram were recorded in every group.Result①The rats appear the abnormal behavior after by penicillin peritoneal injection. It up toⅣorⅤgrade as to Racine scale.②The bone marrow stromal stem cell is cultured achievly.③In the fluorescence microscope, there isn't the fluorescence cells in the brain sections of sodium chloride group. But the cells can be seen in the BMSCs goup. As the time go on, the survival cells were decreasing. The cells in the 2 weeks is less than 1 weeks, 4 weeks is less than 2 weeks (P<0.01) . ④HE staining show: compared with group of the epileptic without treatment and the epileptic with Sodium Chloride, the number and degree of the neuron necrosis and absence obviously decrease in the hippocampus by the BMSCs treatment.⑤Immunohistochemical stain show: compared with group of the epileptic without treatment ,the Bax positive cell ,Bcl-2 positive cell and the rate of Bax/Bcl-2 hasn't obvious difference in the group of the epileptic with Sodium Chloride . Bax positive cell decrease ,Bcl-2 positive cell increase, and the rate of Bax/Bcl-2 decrease in BMSCs group compare with group of the epileptic without treatment. Compared with group of the epileptic with Sodium Chloride in 1 week, 2 weeks, 4 weeks, Bcl-2 positive cell increase, and the rate of Bax/Bcl-2 decrease in BMSCs group in 1 week, 2 weeks and 4 weeks. Compared with BMSCs of 1 week, the Bcl-2 positive cell increase, Bax positive cell decrease and the rate of Bax/Bcl-2 decrease in BMSCs of 2 weeks.Compared with BMSCs of 2 weeks, the Bcl-2 positive cell increase, Bax positive cell decrease and the rate of Bax/Bcl-2 decrease in BMSCs of 4 weeks().⑤Compared with group of the epileptic without treatment, the EEG of group of the epileptic with Sodium Chloride hasn't improvement. Compared with normal control group ,along with the time go on , bone marrow stromal cells transplantation can decrease the frequency of discharge of the epileptic rats, and reduce the amplitude of discharge to half its previous level.Conclusion 1 The epileptic model of the SD rats were established successfully by injection of penicillin at belly. It up toⅣorⅤdegree.2 The bone marrow stomal stem cells has the ability of survival and immigration in the epileptic rats. But the number of the survived cells decreased gradually along with the time go on. 3 The expressing of the bcl-2 protein increased,and the expressing of the bax protein decreased,the value of bax/bcl-2 decreased in the epileptic cells of the hippocampus regions after bone marrow stromal cells transplanting.It indicated the transplantion of bone marrow stromal cells in some certain can depressed the cells apoptosis of the hippocampus regions in epileptic hippocampus.The specific mechanism of action need to be further detected.4 Bone marrow stromal stem cells transplantation can decrease the frequency of discharge of the epileptic rats.It has obvious effect of counteracting epileptic.
