[Objective]:To investigate the expression and significance of NKCC1、KCC2andBDNF in the epileptogenic zone of the focal cortical dysplasias Ⅱa(FCDⅡa) relativeintractable epilepsy.[Methods]:1、The expression of NKCC1、KCC2and BDNF were checked by superSP staining of immunohistochemistry in twelve cases of the epileptogenic zone of theFCDⅡa relative intractable epilepsy and eight cases of normal brain tissue whichsurgical removal of acute intracranial hypertension after traumatic brain injury weretaken as a control group.2、The contents of NKCC1、KCC2and BDNF were checked by Western blotting inone case of FCDⅡa and normal brain tissue respectively.[Results]:1、immunohistochemistry:The expressions of NKCC1、KCC2and BDNFare positive in neurons of control group,However,The expressions of NKCC1、KCC2and BDNF are strong positive in dysmorphic neurons(DNs) of the FCDⅡa.There aresignificant differences in statistics(P value:0.0027、0.0431and0.0061<0.05)。2、Western blotting:The gray value of the FCDⅡa and normal brain tissue isNKCC1226and881;KCC22913and1134; BDNF3225and1013。 The quantification ofNKCC1、KCC2and BDNF of FCDⅡa cell homogenates are more than normal braintissue。[conclusion]:1、NKCC1、KCC2and BDNF may contribute to the development ofepilepsy in FCDⅡa。 2、The increasing of NKCC1and KCC2in the DNs may rusult in the excitability ofGABA,leading to development of epilepsy in the FCDⅡa.3、The BDNF may inhibit the development of epilepsy in the FCDⅡa by controlingthe KCC2of the DNs.
