Objectives:To investigate the expression and distribution of BDNF in Epileptic foci ofFCDⅡa with refractory epilepsy, and to explore the highly expressed BDNF mayrelate to abnormal activation of mTOR signaling pathway in the foci of FCDⅡawith refractory epilepsy.Methods:The Epileptic foci were obtained from Surgical patients with medically intractableepilepsy in our Hospital from January2009to December2010. These Epileptic foci,which pathologically confirmed FCD Ⅱa(n=8)and FCDI(n=16), were took as the twoexperimental groups of our study object. Normal cortex obtained from the temporal lobebrain tissue which is inevitably to be removed in the patient of traumatic brain injury asa control group. We investigated and statistical analyzed the expression and distribution ofBDNF by immunohistochemistry and western blot in each group. By doubleimmunofluorescence staining, We evaluated whether BDNF co-expressed withp-AKT(Ser473) and p-mTOR(Ser2448) in epileptic foci and control group.Results:1. Immunohistochemistry: BDNF was mainly distributed and expressed in neuronsof the FCD Ⅱa type, especially strongly positive in dysmorphic neurons of FCDⅡa type; BDNF was mainly positive in neurons of the FCDⅠ type; maindistribution in the FCDI type; BDNF was moderate intensity expression inneurons of control group.(each group compared, P<0.5)2. Western blot:The amount of BDNF expression in the case of each group:①F CDⅡa group was significantly higher than FCDⅠ group and the controlgroup;②FCDⅠ group was higher than the control group.(each group compared,P<0.5)3. Immunofluorescence double labeling staining: BDNF respectively co-expressed with p-Akt (Ser473) and p-mTOR (Ser2448) in DNs of the FCD Ⅱa type, thecoincidence rate was almost100%, substantially,other cells was not co-expression,p-AKT (Ser473) and p-mTOR (Ser2448) almost not expressed in the FCD Ⅰtypegroup and normal brain tissue.Conclusions:1. BDNF was high expression in epileptic foci of FCDⅡa type with refractory,especially expressed in DNs,Almost not in glial cell, It may be related to theirseizures.2. BDNF may be one of upstream factors of abnormally activated mTOR signalingpathway in DNs of epileptic foci of FCD Ⅱa type with refractory epilepsy.
