Post-traumatic stress disorder (PTSD) is a kind of abnormal psychic recation, whichdevelops after a strong spiritual trauma. With the rapid development of society, the stressevent is growing, the stress intensity is increasing. More and more people suffer fromPTSD. The incidence of PTSD increases rapidly. PTSD can cause significant occupational,psychological and social functional disability. They bring significant economic burden tothe patient himself, their family, and community. Therefore, the prevention of PTSDbecomes important and significative.This study started from the preparation of similar clinical PTSD model, singleprolonged stress (SPS) model. The model mice were immediately given zhizichi decoction,ganmaidazao decoction, dastrodine capsule or physiological saline after SPS treatment. On this basis, we explored the effects of drugs on the prevention of PTSD, and providedscientific evidence for clinical prevention of PTSD.ObjectivesThe model rice were immediately given zhizichi decoction, ganmaidazao decoction,dastrodine capsule or physiological saline after SPS treatment. We evaluate the effects ofdrugs on behavior, neuroendocrine hormones and hippocampal cytokines of rats withPTSD. Then we explore the effects of zhizichi decoction, ganmaidazao decoction,dastrodine capsule on the prevention of PTSD.Materials and methods1. Experimental animal: SPF male SD40rats were randomly divided into5groups:control group (n=8), model group (n=8), zhizichi decoction (ZZCD) group (n=8),ganmaidazao decoction (GMDZD) group (n=8), gastrodin group (n=8).2. Model preparation: First, the mice were detained for2h. Immediately, they wereforced to swim20min. Then they rest for15min. In the end, they were loss ofconsciousness by aether.3. Experimental methods: The animals were given physiological saline or drugs from1stto14thday after SPS treatment. The15thday we do OF test. The MWM test wasobserved from16thto20thday.All rats were killed at the21thday. The levels of IL-1β, IL-6and TNF-α in hippocampal were detected by ELISA. The level of ACTH in plasm wasdetected by RIA. The levels of COR, TSH, T4, T3, LH and testosterone in serum weredetected by RIA.Results1. Results of OF test: Compared with control group, the erect quantity of modelgroup was less (P <0.05). There were no statistically significant in other experimentalindexs(P>0.05).2. Results of MWM test: In comparison with control group, a striking increase of thelatency was observed in model group(P <0.05).3. IL-1β, IL-6, TNF-α content changes: Model group compared with control group,the levels of IL-1β, IL-6in hippocampal increased significantly (P <0.01), the levels of TNF-α in hippocampal increased (P <0.05). ZZCD group compared with model group, thelevels of IL-1β, TNF-α in hippocampal were significantly lower, with a significantdifference (P <001), the levels of IL-6in hippocampal reduced(P <0.05). CMDZD groupcompared with the model group, the levels of TNF-α in the rat hippocampal wassignificantly lower, with a significant difference (P <0.01), the levels of IL-6inhippocampal reduced (P <0.05), the levels of IL-1β in hippocampal reduced, but notstatistically significant (P>0.05). Gastrodin group compared with the model group, thelevels of IL-6, TNF-α in hippocampal changed (P <0.05); the levels of IL-1β inhippocampal were not statistically significant (P>0.05).4. ACTH and COR content changes: The result showed that the levels of ACTH,COR were significantly increase model group in comparison with control group (P <0.01).GMDZD group, ZZCD group and Gastrodine group could descend the contents of ACTHand COR, and there was no significant difference between three drug groups(P>0.05).5. TSH, T4and T3content changes: Model group compared with control group, thelevels of T4in serum increased significantly (P <0.01), the levels of TSH, T3in serumwere higher, but the differences were not statistically significant (P>0.05). Comparedwith model group,the contents of T4and T3in serum, three drugs groups decreasedsignificantly, the difference was statistically significant (P <0.01); the levels of TSH inserum were not statistically significant (P>0.05). There were not statistically significantdifferences among the groups of drugs on the contents of TSH, T4, T3in serum (P>0.05).6. LH and testosterone content changes: The differentces of LH levels in serum werenot statistically significant among5groups (P>0.05). Model group compared with controlgroup, the levels of testosterone in serum were significantly increased, and the differencewas statistically significant (P <0.01). Compared with the model group, the levels of LHin serum in GGDZD group, ZZCD group, and gastrodin were no statistically different (P>0.05), but the levels of testosterone in serum were not statistically significant (P>0.05).Three drugs groups in comparison with control group, the levels of LH and testosterone inserumshowed no significant difference (P>0.05). There were not statistically significantdifferences among the groups of drugs on the contents of LH and testosterone in serum (P >0.05).Conclusions1. Compared with control group,the anxiety-like behavior increased, the spatiallearning and memory ability were impaired after SPS. There were disorders ofinflammatory cytokines in hippocampus of model group. Zhizichi decoction,Ganmaidazao decoction and gastrodin may improve the disorders of inflammatorycytokines in hippocampus.2. In comparison with control group, there were disorders of HPA axis, HPT axis,HPG axis after SPS. Zhizichi decoction, Ganmaidazao Decoction and gastrodin cancorrect these disorders.
