Objective:To study the expression and significance of Sirt3in adipose tissue of rats on dietswith different calories and the relationship between Sirt3and obesity. Try to providethe foundation for the research of obesity related factors, May also be for a newapproach for treatment of obesity in the future.Methods:1、Select60SD male rat (ablactation, individual weight60g) and divide the ratinto3groups randomly: group A is feed with high calorie diets to construct obesitygroup (OB); group B is feed by basal diets freely to create Ad Libitum group (AL);group C is feed by restricted basal diets to make Caloric Restriction group (CR)2、All groups are applied euthanasia after16weeks and extract adipose tissue todetect the Sirt3gene expression.3、Expression of Sirt3in mRNA and protein are examined by Real-time RT-PCRand Western-blot.Results:1、Success in creating the OB group with high calorie diets.2、Western-blot:OB has a lower level of Sirt3in protein of adipose tissue than AL group(P<0.05); CR group has a higher level of Sirt3in protein than AL group (P<0.05).Statistical significance was observed from the data.3、Real-time RT-PCR:OB has a lower level of Sirt3in mRNA of adipose tissue than AL group(P<0.05); CR group has a higher level of Sirt3in mRNA than AL group (P<0.05). Statistical significance was observed from the data.4、Similar test result has been observed in Western blot and Real-time RT PCRtest.Conclusions:1、Construction of SD rats diet-induced obese model by high calorie diets issimple, easy to operation and the blood lipid test will be a significant change, butbecause of the Feed formulation and obesity resistance, the success rate is low.2、Expression level of Sirt3mRNA and protein in adipose tissue of rats on dietswith different calories can appear obvious difference, high calorie diet would reduceits expression, calorie restricted diet can increase the expression.3、Obese SD rats Sirt3mRNA and protein expression level in the adipose tissueis lower than normal SD rat.
