Objective:To evaluate the fluorescence in situ hybridization (FISH) as anon-invasive diagnostic methods in the clinical feasibility of upper urinary tracttumors.Method:In July2011to October2012, Collect voided urine specimens of20healthy volunteers. Urine cell3,7,17chromosome and chromosome9variant by usingFISH detected, the threshold of the normal population was establish, and urinespecimens from23cancer patients of upper urinary tract tumors and21cases ofcontrol group with benign disease, were analyzed by means of urine cytology andFISH tests. The histopathological diagnosis as standard, to compare two kinds oftechnologies of the result, evaluation the feasibility of FISH.Result:There are23cases were proved to be the upper urinary tract urothelialcarcinoma in25suspicious patients with upper urinary tract tumor. Another2case isrenal cell carcinoma infiltrating renal pelvis and ureter fibroma. And there were1caseof stage PTa,3cases of PT1,9cases of PT2,7cases of PT3and3cases of PT4in the23cases of upper urinary tract urothelia carcinoma. Then,for the cell grade,therewere9cases of low grade and14cases of high grade in the23cases of upper urinarytract carcinoma. FISH detection overall sensitivity on urinary tract urothelialcarcinoma was82.61%, and compared with39.13%of urine cytology, the differencehas statistical significance (P=0.006),and similar specificity, were90.48%and95.24%, respectively (P=1.000). The sensitivity of FISH and urine cytology in23patients with upper tract urothelial carcinoma in the low level of the sensitivity oftumors were66.67%and22.22%, respectively, the high level of sensitivity of tumorswere92.86%and50.00%, respectively, and P value is0.153and0.033, respectively.FISH and urine cytology in23patients with upper tract urothelial carcinoma, Thedetection rate of3,7,9chromosome and chromosome17was69.57%,73.91%,47.83%and65.22%, respectively. In the control group, the FISH and urine cytologypositive cases were2cases and1case respectively. Conclusion:The sensitivity of FISH in the upper urinary tract urothelialcarcinoma obviously higher than urine cytology, and their specificity is similar. FISHcan be used as a new noninvasive method for upper urinary tract urothelial carcinomadiagnosis.
