Atractylodes lancea is the clinical common Chinese medicine, there is spleen dampness effect. Atractylodes lancea has two types according to the "Chinese Pharmacopoeia", and there are two decoction pieces:Atractylodes lancea and Bran fried Atractylodes lancea.This thesis is part of the National Natural Science Foundation Project "the processing mechanism of bran fried Atractylodes lancea ". It focuses on the discussion bran fried Atractylodes lancea reducing dryness efficacy and the changes of the material base to carry out a series of studies. This is a continuation of the previous research work.The previous research work has been finished the chloroform efficacy, and analyzed the chemical composition of chloroform with the method of HPLC, found that the contents of chemical constituents has been changed after processing Atractylodes lancea. On this basis, using silica gel column chromatography and Toyopearl-gel column chromatography separated the chloroform, get two monomer compounds, identified the two compounds are5-hydroxymethyl furfural and diisobutyl phthalate.The "damp retention in middle-jiao" rat mode was established. Small intestine peristalsis rate, GAS level were measured to evaluate the Spleen and Stomach Function. Urine volume, and AQP2content from urine, were observed to testify the eliminating dampness effect. According to the results, bran fried Atractylodes lancea had significant difference with negative group (p<0.05). The Atractylodes lancea gastrin level had no significant differences with negative group; Bran Fried Atractylodes lancea has significant difference compared with negative group (p<0.05), each group increased urine volume, decreasing the content of AQP2, the crude group increased significantly, the most close to the normal value.Using immunohistochemical method, study on the effects of volatile oil on the level of AQP2. According to the results, the crude groups had significant difference to the negative control group(p<0.05), Bran fried group had no significant difference to the negative control group.Experimentation is required to determine the lethal dose (LD50) of volatile oil from Rhizoma Atractylodis and bran fried Atractylodes lancea, analysis the impact of acute toxicity study on volatile oil from Atractylodes lancea through processing. With equal proportion consistency intragastric infusion method, calculation the lethal dose (LD5o) of volatile oil from Atractylodes lancea and bran fried Atractylodes lancea using modified Kou Shi method. Measured of the volatile oil from Atractylodes lancea LD50for2454.71mg/kg, LD5095%confidence limits for2123.24mg/kg~2837.92mg/kg; the volatile oil from bran fried Atractylodes lancea LD50for5248.07mg/kg, LD5095%confidence limits for4677.35mg/kg-5888.43mg/kg. The volatile oil from Atractylodes lancea rating is low toxicity; volatile oil from bran fried Atractylodes lancea is actual non-toxic.On the Basis of the effect of Atractylodes lancea volatile oil, using GC-MS method analyse the constituents of the volatile oil before and after bran. To analyse the high content of component of the Atractylodes lancea oil, found that7components had declined after bran,compared with fried wheat bran four new components from the wheat bran of bran fried with Atractylodes lancea arised. Results showed that the partial dry ingredients of Atractylodes lancea are absorbed after bran.Through this experiment, verified the medical effect was promoted, and the side effect concerning the "zao" property of Atractylodes lancea can be alleviated by frying with bran. Meanwhile, this research provided a reference for further quality and quantity changes research.
