Through the survey of Beijing authorities, Gardeniae Fructus, Atractylodes Macrocephala and achyranthes bidentata are commonly used Chinese medicine in the clinical. Chinese Pharmacopoeia (ChP)2010included Gardeniae Fructus, stir-frying Fructus Gardenia, Gardeniae Fructus Preparatus, Atractylodes Macrocephala, Rhizoma Atractylodis Macrocephalae Stir-fried with Bran, the soil-fried of Atractylodes Macrocephala, deep-fried atractylodes macrocephala koidz, achyranthes bidentata and Prepared Twotoothed Achyranthes Root with Wine. The qualitive identification in their quality standards usually used Thin Layer Chromatography (TLC), the content determination of their index components always used chromatography. However, traditional Chinese medicine (TCM) has complicated chemical composition and a variety of processing methods. For traditional Chinese medicine, there are still some deficiencies need to be improved in their quality standards. On the basis of their present quality standards, these herbal pieces were studied by following ways in order to improve the quality control level of their quality standards.1. The establishment of TLC identification by C18TLCThe TLC identification of nine herbal pieces was established by using C10TLC instead of Silica gel TLC for the first time. To compare with that in their quality standards, the C18TLC identification established in this research of Atractylodes Macrocephala and the processed productsacquire more information, the accuracy of identification is also improved. Meanwhile the sample pretreatment method and developing solvent is simpler and safety. The imformation from the C18TLC identification is basically as same as HPLC, it can be used for the rapid test for drugs.2. The study of HPLC fingerprintThe HPLC fingerprints of Gardeniae Fructus, Atractylodes Macrocephala, achyranthes bidentata and theirprocessed productswere established. There are7characteristic peaks in the fingerprint of Gardeniae Fructus and stir-frying Fructus Gardenia, There are5characteristic peaks in the fingerprint of Gardeniae Fructus Preparatus.The similarity of10batches of Gardeniae Fructus was higher than0.95, the similarity of10batches of stir-frying Fructus Gardenia was higher than0.90. the similarity of9batches of stir-frying Gardeniae Fructus Preparatus was higher than0.70. There are10characteristic peaks in the fingerprint of Atractylodes Macrocephala and its processed products, the similarity of different batches of Atractylodes Macrocephala, Rhizoma Atractylodis Macrocephalae Stir-fried with Bran and the soil-fried of Atractylodes Macrocephala was higher than0.95. the similarity of10batches of deep-fried atractylodes macrocephala koidzwas higher than0.70.The HPLC fingerprints of achyranthes bidentata and its processed productwere established dividually and there are14characteristic peaks in the fingerprint of them, the similarity of14batches of achyranthes bidentata was higher than0.90,, the similarity of5batches of repared Twotoothed Achyranthes Root with Wine was higher than0.70.The HPLC-ESI-MS/MS was used to identify the chemical constituent of characteristic peaks in the fingerprints. The chemical constituent of8characteristic peaks of the fingerprint of Gardeniae Fructus and its processed productswere identified. There are5characteristic peaks of the fingerprint of Atractylodes Macrocephala and the processed products were identified. The chemical constituent of7characteristic peaks of the fingerprint of achyranthes bidentata and Prepared Twotoothed Achyranthes Root with Wine were identified. The identification of characteristic perks made the information from the fingerprints more abundant and improved the level of quality standards, it lays a foundation for further study.3. The content determination of multi-target ingredientsThe method for content determination of genipin-1-β-D-gentiobioside,geniposide and crocin-I in Gardeniae Fructus and the processed products by HPLC was established. The method for content determination of Atractylenolide Ⅰ, Ⅱ,Ⅲ and Atractylone in Atractylodes Macrocephala and its processed products by HPLC was established. The method for content determination of β-Ecdysonein in achyranthes bidentata and Prepared Twotoothed Achyranthes Root with Wine by HPLC was established. These methods are precise, stable, accurate and good reproducibility for the quality control.4. The study of water decoctionThe method of content determination of the water decoction of Gardeniae Fructus, Atractylodes Macrocephala and achyranthes bidentata was be established.The content variation of genipin-1-β-D-gentiobioside, geniposide and crocin-I from Gardeniae Fructus and its processed productss in decoction is studied. The content variation of Atractylenolide I,11,111and Atractylone in Atractylodes Macrocephala and the processed products in decoction is studied. The content variation of β-Ecdysonein from achyranthes bidentata and Prepared Twotoothed Achyranthes Root with Wine in decoction is studied. The results show that, the content of chemicals in medical materials has a considerable change by using different solvent and extraction method, and the change has a certain change rule.
