Objected:To investigate the effect of microRNA-155(miR-155) over-expression on proliferation,apoptosis and migration in laryngeal squamous cell carcinoma Hep2cells.Method:miR-155mimics were transfected into Hep2cells by Lipofectamine RNAi MAX. miR-155expression level was detected by Real-time PCR. Cell proliferation were evaluated by CCK-8assay and colony formation. Cell apoptosis were analyzed by flow cytometry and Hoechst33342and propidium iodide staining. Cell migration were detected by wound healing assay.Result:After transfection of miR-155mimics into Hep-2cells, the data of qRT-PCR showed that miR-155expression level was significantly up-regulated. CCK-8assay and colony formation revealed that growth or proliferation ability of miR-155group was obviously promoted.Cell apoptosis or death was suppressed after over-expressed miR-155which was analyzed by flow cytometry and Hoechst33342&propidium iodide staining. Remarkable enhancement of cell migration was observed in miR-155group which was detected by wound healing assay.Conclusion:Over-expression of miR-155could induce proliferation and migration of Hep2cell line, and inhibit apoptosis. It might play the function as an oncogene in the development of laryngeal squamous cell carcinoma.
