Objective:Detection of miRNA-21expression in laryngeal cancer tissues and cells. To construct the antisense of hsa-miR-21lentiviral vector to observe the impact of recombinant lentivirus of antisense oligonucleotides targeting regulatory Ras gene on laryngeal carcinoma cell proliferation, cell cycle, apoptosis and inhibition of invasion.Methods:Detection of miR-21expression in laryngeal cancer tissues and cells, using real-time quantitative PCR. Constructed antisense of hsa-miR-21lentiviral expression vector, in vitro transfection of laryngeal squamous cell carcinoma Hep-2cells by MTT assay and colony formation assay. The detection of antisense oligonucleotides of miR-21(ASO-miR-21) recombinant lentivirus of laryngeal carcinoma cell proliferative capacity. Detection of laryngeal squamous cell carcinoma cell cycle and apoptosis by flow cytometry. Boyden, chambers to detect the invasion of the laryngeal squamous cell capacity. Western blot detection of RAS protein expression in laryngeal squamous cell carcinoma.Results:PCR detection in laryngeal carcinoma tissue and cell of miRNA-21expression was significantly increased with clinical and pathological staging and grading related. Successfully constructed and the antisense of hsa-miR-21of the lentiviral expression vector, the ASO-miR-21lentiviral to inhibit the high expression of miR-21activity in laryngeal squamous cell carcinoma, can significantly inhibit the proliferation of laryngeal carcinoma cells, inducing apoptosis and arrest Hep-2cell cycle at G1phase and inhibit their invasion. Western blot analysis of laryngeal squamous cell carcinoma cells transfected with ASO-miR-21after Ras protein expression were markedly reduced.Conclusions:This study found that laryngeal carcinoma tissue and cell of miRNA-21expression significantly increased transfection ASO-miRNA-21can inhibit the laryngeal cancer cell proliferation and induce apoptosis and arrest of Hep-2cell cycle at G1phase, and inhibiting invasion. In order to explain the role of miR-21is to play the carcinogenic, a valid target and the theoretical basis for gene therapy for laryngeal cancer and other malignancies.
