Objective Osteoarthritis is a common chronic joint disease, there are manytreatment methods,and are based on conservative treatment.However, these treatmenthas been a difficult clinical problem,is not fundamentally prevent the pathologicalprocess of osteoarthritis.In recent years, the medical profession are actively exploringnew therapies and new technologies for the treatment of osteoarthritis.The role ofintra-articular injection of drug therapy has already been proved by years of clinicalpractice, a large number of patients benefit.This paper studies New Zealand whiterabbits, by which establishes Models of Osteoarthritis, application of5-Flurouracil(5Fu)injecting in joint treatment of rabbit osteoarthritis,by general observation, electronmicroscopy and serum determination of joint synovial fluid interleukin-1β (IL-1β) andsuperoxide dismutase (SOD),explores the mechanism of5Fu in treating osteoarthritis byinjecting in joint.Provide the experimental basis for clinical treatment.Methods Choosing24healthy and clean New Zealand white rabbits in accordancewith the experiment, weighing2.0kg-2.5kg, age six months and adult male. Applicationrandom number table,the experimental animals were divided into5-Fluorouracilgroup(5Fu group),sodium hyaluronate group(SH group) and saline group(control group)(n=8).At1st,4th and7th day,fixed limbs of experimental animals,edge midpoint intoneedle aspiration into the joint cavity with5ml syringe,withdrawing without blood,to4%papain0.3ml injected into the rabbit knee joint cavity,after one week we can get therabbit osteoarthritis model. In accordance with the experimental groups,respectively,5-fluorouracil, sodium hyaluronate, and normal saline injecting in joint.Once a week forfour consecutive weeks, one week after the last injection,sacrificed the experimental animals.By general observation of joint synovium, cartilage and joint synovium electronmicroscope.Collecting Interleukin1β(IL-1β) in the serum and synovial fluid by usingdouble antibody sandwich method to make it react. By reading OD from the position on450nm of Micro-plate Reader and drawing the standard curve, compute theconcentration of IL-lβ. Measure the Superoxide Dismutase (SOD) concentration ofserum and joint fluid by the method of Xanthine oxidase.Results (1)General observation:5Fu group synovial thicken slightly,cartilagetissue without obvious defects,articular cartilage surface roughness and coulorgray.Compare with5Fu group,SH group and control group synovial proliferateobviously,articular cartilage surface showing dark color and ulcer formation.Controlgroup had the most significant of the synovial thicken, part of the cartilage defect andshedding,it can be seen subchondral bone(.2)Electron microscope:The normal synovialcells can be observed in normal group, cell membrane is complete.The pyknotic cell canbe observed in5-Fluorouracil group, the electronic density of the nucleus is higher,Many spines may be observed around the nucleus, the apoptosis had formed. Part of themitochondrias express vacuolar denaturation in SH group, lysosomals and roughendoplasmic reticulum can be observed in the cytoplasm. Many mitochondrias expressvacuolar denaturation in control group and there is not normal cell structure.(3)IL-1βcontent in5Fu treatment group is lower than Sodium hyaluronate group and controlgroup(the differences are statistically significant)(P<0.01).(4)SOD content in5Futreatment group is higher than Sodium hyaluronate group and control group(thedifferences are statistically significant)(P<0.01).Conclusion5Fu joint injection can lower the IL-1β content of rabbit after theosteoarthritis and stimulate the SOD, therefore lessen the joint soft tissue injures andspeed up the recovery of joint. This experiment shows5Fu joint injection has the effectof lessening inflammatory reaction and stimulating the recovery of joint softtissue.Provide the basis for5Fu in clinical treatment of promotion and use.
