Objective To analyze the chronic viral hepatitis fibrosis treated with traditional Chinese medicine by Meta, and then rat experimental study of intersection herbs from Meta-analysis was performed to observe the preventive effect on liver fibrosis.Methods Extensive retrieved literature search for human studies were performed, in near36years up to May in2012. MEDLINE and Cochrane Controlled Trails Register (CCTR), Chinese science and technology journal full-text database, Index included Chinese National Knowledge Infrastructure (CNKI), China Biomedicine Literature Database (CBM), The standard of inclusion and exclusion were formulated. Quality assessment of the literatures of randomized clinical trials (RCT) used the four rules of Cochrane Handbook and Jadad’s score. The whole effect of RCTs was described by RevMan5.1software, and funnel plot was used to watch the potential publication bias. Intersected herbs were acquired from Above Prescriptions of RCTs based on Meta-analysis. Rats were divided into several groups by randomization which included model group, normal control group and the intersection of traditional Chinese medicine (single and compound) group. Liver fibrosis in rats were induced by Carbon tetrachloride (CCL4) injections for6weeks, at the same time, treated with intersected herbs. Liver HE staining and Masson staining were performed. The fibrosis extent of liver Massons stain was assessed by IPP software with Area and integral optical density(IOD). Results Six studies were consistent with the inclusion criteria, six effective prescriptions were analyzed and set. There were significant differences between therapeutic groups and control groups, and its total RR=2.29(95%CI1.76-2.97, Z=6.19, P<0.00001). Intersection of herbs of six prescriptions included Salvia and Astragalus. Compared to Salvia group and Astragalus group, the rats liver tissue of model group were obviously damaged, and their collagen fibers were increased. The Salvia+Astragalus group was not damaged seriously, and their staging of liver fibrosis was obviously improved, the Area and IOD of collagen fibers were obviously decreased (p<0.01). Collagen area and IOD in Salvia group or Astragalus group was lower than that in model group (P<0.01), while the stage of liver fibrosis, collagen area and integral optical density IOD in Salvia+Astragalus group was significantly lower than that in Salvia group or Astragalus group (P<0.01).Conclusion Compound traditional Chinese medicine on the prevention of hepatic fibrosis has obvious effect than others. Salvia and Astragalus could prevent the expression of liver fibrosis in rat. Salvia combined with Astragalus is better than single drug.
