ObjectiveThe relationship between serum folate,RBC folate, SAM, DNMT1as well as MeCP2proteins was mainly investigated on the condition that association between risk factors of cervical cancer and cervical cancer so as to provide a theoretical evidence for the study of etiology and mechanism of cervical cancer.Methods54cases with squamous cervical cancer,43cases with CIN Ⅰ,44cases with CIN Ⅱ-Ⅲ and75cases with cercitis were selected from Tumor Hospital affiliating to Shanxi medical University from June, Health Care Center for Women and Children of Taiyuan and Health Care Center for Women and Children of Jie Xiu from2009to2011.All sujects got a definite diagnosis through pathological exam, did not receive radiotherapy and chemotherapy before we gained biological material from them. Those sujects with megaloblastic anemia, haemolytic diseases, leukemia,local enteritis, live diseases and other types of tumers were excluded. and those taking in Vitamin B within three monthes were also not included.Information about the demograthy, reproductive, life habits et al were colleced through structural questionair. At admission, we get the tissues of cervix. Microbiological assay was adopted to test the level of serum folate and RBC folate, HPLP to SAM level of whole blood, and Western-blot to test MeCP2and DNMT1protein expression level. We established database and analyzed data by using SPSS16.0. Wilcoxon order Sum and ANOVA were used to analyzed the qualitative data,χ2Test and χ2Trend Test to analyze the categorical data, and Logistic Regression to analyze the risk factors of cervical cancer.Results1) Difference of levels of serum folate and red blood cell folate in different groups by the Wilcoxon rank sum test was significant(Z=59.41, P<0.001; Z=15.63, P=0.001) and with the worse of cervical cancer, the levels of serum folate gradually reduced with significance (χ2trend=51.222, P=0.001;χ2trend=9.793, P=0.002)2) Difference of levels of SAM in different groups by the Wilcoxon rank sum test was not significant(Z=3.51, P=0.319) and with the worse of cervical cancer, the levels of red cell folate gradually reduced with no significance but near significance (χ2trend=3.838, P=0.050).3) Difference in protein expression levels of DNMT1and MeCP2in different groups by the ANOVA variance analysis was significant(F=21.576, P=0.001; F=16.088, P=0.001) and with the worse of cervical cancer, the levels of levels of DNMT1protein expression gradually increased with significance(χ2trend=9.793, P=0.002;χ2trend=41.555, P=0.001).4) Results of correlation analysis showed that posive correlations between serum folate and red blood cell folate with significance (r=0.30, P=0.001), between red blood cell folate level and SAM level with non-statistically significant (r=0.066, P=0.377), between NDMT1protein expression levels of MeCP2protein expression levels,as well as between DNMT1protein expression levels and MeCP2protein expression levels with statistically significant (r=0.299,P=0.001); and negative correlations existed between between serum folate level and SAM level with non-statistically significant (r=-0.125,P=0.067),between serum folate level andDNMT1protein expression levels with statistically significant(r=-0.270,P=0.001), between red blood cell folate level and DNMT1 protein expression levels with statistically significant (r=-0.199, P=0.080), between serum folate level and MeCP2protein expression levels with statistically significant(r=-0.268, P=0.001), between red blood cell folate level and MeCP2protein expression levels with statistically significant(r=-0.110, P=0.106).6) Results of the categorial non-conditional Logistic regression analysis showed that four factors of places of residence,gynecological history,family history of cancer and DNMT1protein expression entered CIN Ⅰ Logistic regression, that eight factors of place of residence, frequency of washing vagina, parity, gynecological history, family history of cancer, serum folate, DNMT1protein levels and MeCP2protein expression levels entered CINⅡ-Ⅲ Logistic regression and that seven factors of age, occupation, frequency of washing vagina, gynecological diseases, serum folate, DNMT1protein expression levels and MeCP2protein expression levels entered Cervical cancer Logistic regression.Conclusion1) deficiency of Serum folate and RBC folate are risk factors for cervical cancer.2) With the severity of cervical lesions, SAM levels decreased, but this trend was not significant, and at the significant level significantly.3) Fact that with the worse of cervical cancer, the levels of DNMT1and MeCP2protein expression gradually increased may lead to the propose that DNMT1and MeCP2protein expression levels may be used as the biological markers of gradually deteriation of cervical cells.4) Place of residence in Shanxi, low serum folate levels, low frequency of washing vagina, high levels of DNMT1protein expressionand high levels of MeCP2protein expression are common risk factors for CIN and cervical cancer, while occupation of farmers is cervical cancer risk factors. Gynecological diseases is the common protective factor for CIN and cervical cancer.
