Objective:In this part we want to acquire the proliferation mechanism regulated by c-Myc about the abnormal expression of NS in prostate cancer cell.Methods:The prostate cancer PC3cell lines weretreated with High-c-Myc-express-ion plasmid,small-molecule c-Myc inhibitor10058-F4,hydrogen peroxide and catalase,subsequently,fluorescent probes、fluorescent microscopeandFluorence microplate reader were used to detect the expression levels of ROS in prostate cancer cell lines.Western Blot was used to detect the expression levels of the NS.MTT and cell counting was used to detect the proliferation in prostate cancer cell,SPSS17.0statistical software was applied to analyze the data and statistical significance was defined as P values less than0.05.Results:C-Myc can regulate the expression of the NS on the level of protein in prostate cancer. The expression of c-Myc can affect ROS and NS, the high expression of c-Myc can promote the expression of the NS and endogenous ROS,while promoting the proliferation of prostate cancer PC3cells. High expression of H2O2can lead to high expression of the NS and promote the proliferation of PC3cells.Catalase can reduce the expression of NS and ROS which is caused by the high expression level of c-Myc,while inhibiting the proliferation of prostate cancer PC3cells (P<0.05).Conclusions:It suggested that the c-Myc-ROS-NS pathway may exist in the prostate cancer, which involved in the proliferation mechanism about the abnormal expression of NS in prostate cancer cell,and we believe that inhibition of c-Myc and its stimulated signaling might prove to be a promising strategy for restraining tumor progression in tumors.
