Research On The Relationship Of GFAP And SNPrs2070935Polymorphism In First Episode Schizophrenia

Objective To study the relevance between serum glial fibrillary acidic protein levels and GFAP SNPrs2070935polymorphism in first episode schizophrenic patients.Methods58patients with first episode schizophrenia were sampled from outpatients and52age-and sex-matched controls were sampled from health examination center. Assessed mental disorders with a self-designed general situation questionnatire and given the scores of Positive and Nega—tive Syndrome Scale(PANSS). Enzy me linked immunosorbent assay (ELISA) were used to evaluate the GFAP concentration in serum. GFAP genetype for polymorphisms of SNPrs2070935were performed by PCR and direct sequencing. T-text, Chi-sequare text (y2text), one-way analysis of variance and spearman correlation were used for data analysis and statistics.Results (1)Serum GFAP lever in first episode schizophrenic were significantly higher than normal controls(4.211±0.72ng/L vs3.33±0.85ng/L, t=5.90,P<0.01). The GFAP serum concentration with different sex, age, course of disease, and score of PANSS were analysed, respectively. The results showed that serum GFAP lever in these elements have no significant difference.(2)The proportions of three genotypes::CC(56.36%)、CA(34.55%、AA(9.09%). In case group, the proportion is44.83%,41.38%,13.79%, respectively; In control group, the proportion is69.23%,26.92%,3.85%, respectively. three genetype had sense in statistics between the case group (x2=7.54, P=0.023), and the normal controls group(χ2=8.33, P=0.004), and three allele frequency had sense in statistics.too (P<0.01).(3)serum GFAP level had no sense in statistics in three genetype (F=0.85, P=0.43), The GFAP serum concentration in different genotype had no sense in case group and normal controls group,too(F=0.73and0.07, P=0.69and0.94).Conclusions Serum GFAP lever in first episode schizophrenic were higher than normal controls, but there is no relationship between GFAP polymerphism and Serum GFAP levels, which show the first episode schizophrenic patients exist glial cell damage. We consider that, under a protective feedback mechanism, GFAP participates in repair and regeneration of glial cell, there is no relationship between GFAP polymerphism and serum GFAP levels, the chang of serum GFAP levels is possibl relate to other factors; On the other hand, the study show that there is a relationship between GFAP polymerphism and first episode schizophrenic,we consider that GFAP polymorphism is possible the danger facter,which lend to schizophrenic.
