Objective:Through the cell migration in vitro experiment and whole animal experiment, verify the influence of directional transplantation of vascular endothelial growth factor to Bone marrow mesenchymal stem cells. Through echocardiography contrast cardiac function before and after stem cell transplantation therapy of animal with myocardial infarction to test whether stem cell transplantation combined with VEGF can enhance curative effect of cardiac function. Provide basic research for clinical use of BMMSCs for myocardial infarction.Methods:Cellular level research irrigating robbit’s bone marrow collect bone marrow cells, cultured primary BMMSCs using bone marrow adherent separation method. Use transwell for migration experiment, the cell can be divided into the experimental group and control group, each group with five transwell (n=5), the experimental group uses the VEGF of factors of chemotactic,BMMSCs inoculation in transwell,after48h remove the transwell, crystal violet staining, microscope count thenumber of migrated cells.Whole animal level research:40healthy robbits were randomly devided into4groups,each group have10animal(n=10). Group A is sham-operation group,opening the chest of animals,but not ligating left anterior descending branch;group B is control group,ligating left anterior descending branch,but not injecting BMMSCs;group C is stem cell transplantation group, ligating left anterior descending branch,and injecting BMMSCs;group D is stem cell transplantation with VEGF group, ligating left anterior descending branch,and injecting VEGF into the edge of myocardia of Infarction,then injecting BMMSCs.The heart infarction model were established with ligating left anterior descending branch. After myocardial infarction1week, A C D group injecting BMMSCs with CM-Dil, after1day put to death animals,taking out the heart,making slices, counting BMMSCs under fluorescence microscope, compared three groups of animals Number of BMMSCs myocardial homing. B, C, D group do echocardiography before myocardial infarction,after stem cell transplantation therapy,before stem cell transplantation therapy,getting EF, LVDd, an LVDs value. Compared three groups of animals changes in cardiac function before and after stem cell transplantation therapy.Results:BMMSCs in vitro migration assay, microscope counting, statistical analysis showed that the experimental group the number of migrating cells significantly more than the control group, the results were significantly different (P<0.05). Through the whole animal experiments, fluorescence microscopy count myocardial homing cell mass, statistical analysis of group C and group D homing cells more than the number of A-group difference was statistically significant (P<0.05); C, D two groups homing no significant difference (P>0.05) in the number of cells.3confirmed by the echocardiography by stem cell therapy to improve cardiac function after myocardial infarction role (P<0.05), and Group D to improve the most obvious effect with B, C group compared to the results difference (P<0.05).Conclusions:①in vitro migration assay, VEGFcan influence BMMSCs directional migration;②in the whole animal experiments, VEGF does not promote the BMMSCs’s homing3. BMMSCs transplationg treatment can improve heart function in animals wit myocardial infarction, if the combination VEGF further increase the therapeutic effect of the improvement of cardiac function.
