[Objective]:By cetuximab down-regulation human small cell lung cancer cells NCI-H209neuroendocrine differentiation (neuroendocrine differentition, NED),to discuss in the role of cetuximab, the change characteristics of NCI-H209small cell lung cancer cell’s neuroendocrine differentiation, proliferation and apoptosis,the possible regulatory mechanisms and relevant factors, in order to study blocking of tumor neuroendocrine mechanism and provide an experimental basis for clinical application.[Methods]:Cell morphology was observed by HE dying, SCLC NCI-H209with cetuximab which before and after co-culture the expression characteristics of CgA, Syn, Bcl-2, Ki-67, TFF-land E-cad was observed by immunohistochemical staining. Cetuximab and the cell growth curve of NCI-H209cell which was cultivated all before and after by CCK-8method.The cell cycle changes of NCI-H209was observed by Flow cytometry and the variation of NCI-H209cell invasion was observed by Transwell cell invasion experiment.[Results]:(1)Before cetuximab treatment,the cancer cells grew single suspension, or tumor cells forming loose ball, irregular shape; after medication, the cells gradually became a dense ball,Training to28days,Control group and experimental group difference was decreased on expression product of IM Syn, Ki-67,Bcl-2and E-cad (P <0.05), CgA and TTF-lexpression product was no obvious change. (2) To detect the cell vitality after cetuximab with NCI-H209cell co-culture24h,48h,72h by CCK-8.After48and72h,the difference between control group and experimental group1、2、3were statistical significance (P<0.05),except experimental group2and3were no statistical significance (P>0.05), other two groups were statistical significance (P<0.05); After48h,experimental group3and other groups were remarkable statistical significance(P<0.01).(3) The culture of NCI-H209cell were played role in by cetuximab.The proportion of cells in G1increased, G2and S decreased by FCM in G1phase difference between control group and experimental group1were statistical significance (P<0.05), and differences between experimental group2,3were remarkable statistical significance (P<0.01);in S and G2. in addition to difference between experimental group2and3were statistical significance (P<0.05), The rest of the group differences were remarkable statistical significance (P<0.01).(4)The number of transmembrane cell cells in the experimental group were (12.1±4.3,12.7±3.2,11.7±3.6,), the difference were between statistical significance group and the experimental group (P<0.05), the difference were no stastical significance between the experimental groups (P>0.05).[Conclusion]:(1)The expression of Syn decreased after NCI-H209cell being worked by Cetuximab.It suggest Cetuximab plays a certain role in down-regulating NED. Cetuximab had the time and concentration dependence when worked on NCI-H209cell.(2) The expression of Ki-67, Bcl-2decreased after NCI-H209cell being worked by Cetuximab after it being worked by cetuximab, the cell cycle was blocked,the proportion of cells in G1phase increased, with S, G2cell ratio decreased.Proliferation ability and anti-apoptosis ability were decreased.(3) The expression of E-cad Iincreased after NCI-H209cell being worked by Cetuxima,the ability of invasion of NCI-H209cells dropped.
