| ObjectiveThrough immunohistochemical (IHC) methods, detection the expression of MGMT and XRCC1in different grades of glioma tissue and normal brain tissues. And analyzes it’s the expression and correlation in different grades of glioma tissue and normal brain tissues. Provide a theoretical basis for the formulation of glioma clinical individual treatment.MethodsBy immunohistochemical technique, respectively detection the expression of MGMT and XRCC1in47cases of different grades of glioma tissue and10cases of normal human brain tissue. Analysis the expression and correlation of them by the statistical method in different grades of glioma tissueã€normal brain tissue and each other and the gender and age.Results1.22patients with gliomas of47were positive with MGMT. The overall positive expression rate is46.81%. The positive rate of expression of different grades of glioma as follows:the positive rate of grade I glioma was55.56%in (5/9);The positive rate of grade â…¡ glioma was41.67%(5/12);The positive rate of grade â…¢ glioma was42.86%(6/14); The positive rate of grade IV was50.00%(6/12).MGMT expression rate in gradea â… and â…£ are higher than grade â…¡ and â…¢. By statistical analysis,the different of different grades of glioma was no statistical significance(P>0.05) 2.18patients with gliomas of47were positive with XRCC1. The overall positive expression rate is38.30%; The positive rate of expression of different grades of glioma as follows:the positive rate of grade â… glioma was33.33%(3/9); The positive rate of Grade â…¡ glioma was25.00%(3/12); The positive rate of Grade â…¢ glioma was50.00%(7/14);The positive rate of grade â…£ was41.67%(5/12). XRRCC1expression rate in grade â… to â…¡ of glioma is lower than gradeâ…¢-â…£. By statistical analysis, the different of different grades of glioma was no statistically significant (P>0.05)3.10cases of cerebral hemorrhage in the resection line decompression surgery within the normal brain tissues of MGMT and XRCC1expression was negative or weakly positive. The expression of MGMT and XRCC1were significantly different compare with glioma tissue (P<0.05).4. MGMT and XRCC1expression in glioma cells was not correlated. There were no significant difference with gender and age of the expression of MGMT and XRCC1in the47patients’glioma organization.Conclusions1. MGMT expression in gliomas of different grades was of different, its expression rate in grade â… and â…£ is higher than gradew â…¡ and â…¢. But statistically analyzes the differences was not statistically significant, explain MGMT expression and tumor grade had no correlation.2. XRCC1expression in gliomas of different grades was of different, its expression rate in grade â… to â…¡ of glioma is lower than gradeâ…¢-â…£.But, statistically analyzes the differences was not statistically significant,explain XRCC1expression and tumor grade had no correlation.3. In glioma, the expressions of MGMT and XRCC1were no relative with age and sex.In clinical diagnosis and treatment process, age and sex can not be used as a reference factor when Setting individualized chemotherapy for patients. 4. MGMT and XRCC1expression in glioma cells were not correlated, this suggesting that two gene in gliomas occurrence and development had not play a mutual adjustment.But can be detected by MGMT and XRCC1to judge the prognosis of glioma patients and formulate individualized treatment plan more reasonable.5. The expression of MGMT and XRCC1in normal brain tissues were negative or weak positive, with a significant difference between the expression of different grades of glioma. |
