Objective to investigate whether platelet-rich plasma gel could promote the healing ofgraft-bone interface after anterior cruciate ligament (ACL) reconstruction.Methods Collectting the fresh platelet-rich plasma at the beginning8hour ofsurgery. Forty-eight New Zealand white rabbits were utilized and divided into threegroups randomly. Using a long digital extensor tendon graft to reconstuct the anteriorcruciate ligament.For the group A, injected with platelet-rich plasma gel. For the groupB, injected with fibrin glue.For the group C, we do nothing. Four rabbits from eachgroup were slaughtered at2weeks,4weeks,8weeks and12weeks,then evaluated byanatomy studies and the patholody dyes such as hematoxylin and eosin、toluidineblue and modified trichrome staining. The number of fibroblasts were counted attendon-bone interface with HE staining. And we obtained the Buark score in threegroups.All of those were in order to investigate whether platelet-rich plasma gel couldpromote the healing of graft-bone interface after anterior cruciate ligament (ACL)reconstruction.Results (1)①G roup A:loose tissue and fibroblasts were found at the tendon-boneinterface at two weeks after the surgery. At the same time,the interface was stillseperated.At4weeks, a large number of loose tissue and fibroblasts could befound.There were a few collagen fibers and chondrocyte-like cells. The interface cleftcould not be found easily. The fresh bone was appeared. At8weeks, large areas ofcollagen fibers and chondrocyte-like cells could be found at tendon-bone interface. Sharpey fiber appeared in bundle shape. At12weeks,dense connective tissues formed.A large number of regular Sharpey fibers and fibrocartilages were found.The calcifiedcartilage could be found at tendon-bone interface.②G roup B、C:at2weeks,few loosetissue and fibroblasts were found at the tendon-bone interface. The interface was stillseperated.Fibroblasts could be found but there were no collagen fibers. At4weeks, alittle loose tissue and fresh bone were found attendon-bone interface. There were only a little chondrocyte-like cells and collagenfibers. At8weeks, There were a few collagen fibers and chondrocyte-like cells.Occasional perpendicular collagen fibers were observed crossing this area.At12weeks,large amount of disorganised chondrocyte-like cells and irregular Sharpey fiber werefound.(2) With HE staining under high magnification microscope,the number offibroblasts were counted at tendon-bone interface.We adopt one-way ANOVA andSNK test to analyze the data.The difference between group Aand group B、group A andC were considered significan(tP<0.05).The comparison between group B and groupC were not obvious.(3) Based on the fibrovascular tissues and Sharpey fiber at thetendon-bone interface, The data of Buark score was analysed with Wilcoxon rank sumtest.Between group A and group B、group A and group C,the Buark scores weresignifcantly different(P<0.05). It was concluded that the Buark score of group A wasmore than group B、C.Conclusion Platelet-rich plasma gel could promote the healing of graft-bone interfaceafter anterior cruciate ligament (ACL) reconstruction.
