Effect Of CIK Adoptive Immunotherapy On The Quality Of Life In Patients With Cancer

Objective: To observe the effect of CIK adoptive immunotherapy on the life quality ofpatients with cancer.Methods:Fifty cases with malignant neotlasm were enrolled in our study confirmed bypathology, imaging and cytology from July2011to July2012. Decimation autologousperipheral50ml, we used Ficoll to separae and collect the mononuclear cells, and thenwashed the mononuclear cells3times with sterile saline. Last we suspended the cellsin RPMI-1640complete medium at2×106/mL. According to bone marrow transplantcenters of the Stanford University, we cultured the mononuclear cells. We added IFN–γ500U/mL to the RPMI-1640complete medium in the first day and cultured the cellsat37°C,5%CO2incubator for24hours, added IL-2300U/mL, CD3l0ng/mL,IL-1100U/mL in the second day. Cell growth was observed every day according tothe conditions for amplification. Medium was changed every three days adjusting thecell number of2×106/mL. Culturing the cells for10to15days,we counted the cellsusing trypan blue counting. After the bacteria, mycoplasma, chlamydia, fungi, andendotoxin testing were negative, we harvested the cells for treatment. Taking theamplified cells and analyzing by flow cytometry on the3th,7th,15th day.Results: After the treatment of CIK, the quality of life in patients improved greatly.The ECOG score, KPS score and QOL score improved to varying degrees withstatistical significance. The appetite, physical, sleep, pain and other conditions were improved too. We analysed the cells by the Flow cytometry. The ratio ofCD3+,CD3+CD56+cells was increased significantly,and the the CD4+CD25+regulating T cells (Tregs) reduced statistically significant after treament.Conclusion:(1) CIK immunotherapy can improve the quality of life and immunity inpatients with cancer.(2) CIK biological therapy combined with chemotherapy caneffectively reduce the adverse effects of chemotherapy, while increasing the efficacy ofchemotherapy.(3) The CIK cells are autologous cell,,thus CIK transfusion process arefew and mild adverse reactions, so treatment is safe.(4) CIK Immunotherapy could bethe forth adjunct treatment following surgery, radiotherapy, chemotherapy.
