OBJECTIVEFrom the differential expression of immunohistochemical detection of neurotrophinfamily members of BDNF, GDNF, NT-4in pancreatic cancer tissue to discussion on:1.The correlation of the neurotrophins family members of BDNF, GDNF, NT-4inperineural invasion of pancreatic ductal adenocarcinoma.2. The effect of BDNF, GDNF, NT-4in perineural invasion of pancreatic ductaladenocarcinoma.METHODS1. Collect20cases with neural invasion of pancreatic ductal adenocarcinoma in group A,10cases of gastric adenocarcinoma without neural invasion in groupB and10casesnormal pancreatic tissue in group C, prepared by conventional histological sections.2. To establish reliable immunohistochemical positive control and negative control, byusing immunohistochemical technique respectively in3groups of tissues were stainedwith immunohistochemistry.3. Detection of neurotrophin family members of BDNF, GDNF, NT-4in pancreaticductal adenocarcinoma tissue and control tissue expression, analysis and statistical processing of the results.RESULTS1. BDNF, NT-4, GDNF were not expressed in pancreatic acinar cells and duct cells innormal pancreatic tissue (group C). NT-4, BDNF were positive expression in pancreaticislet cells, but GDNF were negative or weakly positive expression.2. In group A (Pancreatic ductal adenocarcinoma with perineural invasion phenomenon),cancer cells were diffusely positive expression by BDNF, NT-4and GDNF. Theexpression rate was100%, but the intensity range of three is difference, the expressionof GDNF is weaker than BDNF, NT-4.3. The20cases of pancreatic ductal adenocarcinoma perineural invasion (group A) wereexpressed BDNF, NT-4, GDNF. But expression is no correlation with patient age, tumorlocation, histological type and clinical stage (P>0.05).4. BDNF, NT-4, GDNF in gastric adenocarcinoma tissues without neural invasion(group B) was not expressed.5. BDNF, NT-4, GDNF was expressed in pancreatic ductal adenocarcinoma groupperineural invasion (group A).The expression rate were almost100%.The cases ofgastric adenocarcinoma without neural invasion (group B) were not expressed.Group Aand Group B have obvious differences.CONCLUSION1. There is a correlation between the BDNF, NT-4and GDNF secretion from thepancreatic ductal adenocarcinoma cells with the behavior of neural invasion inpancreatic ductal adenocarcinoma.2. BDNF, NT-4and GDNF in pancreatic ductal adenocarcinoma produced by at leasttwo ways, from autocrine by neural tissue and from paracrine by pancreatic ductaladenocarcinoma cells.The BDNF, NT-4, GDNF secretion from pancreatic ductal adenocarcinoma cells is correlated with perineural invasion in pancreatic ductaladenocarcinoma.3. The BDNF, NT-4, GDNF generated by all two kinds of secretory patterns maypromote the perineural invasion of pancreatic ductal adenocarcinoma cells.4. The main mechanism of BDNF, GDNF, NT-4promotes the neural invasion inpancreatic ductal adenocarcinoma may be relate with the role of nerve tissuechemotactic induction and regeneration by BDNF, GDNF, NT-4which secretion fromcancer cells...
