ObjectiveStudy on the relationship between the refractory function of BDNF and GDNF levels in patients with refractory obsessive-compulsive disorder, Established the rat model of obseesine-compulsive disorder, observed the rat behavior change, BDNF and GDNF in the blood of rats were measured, and also measured dopamine content in rat brain tissue,the changes provide a reference for the study of the pathogenesis of refractory obsessive-compulsive disorder. Methods 1. The cognitive function of 41 patients with refractory obsessive compulsive disorder was assessed using the neuropsychological assessment tool(RBANS). 2. Determination of BDNF and GDNF in blood of patients with obsessive-compulsive disorder by ELISA. 3. 16 adult healthy male SD rats were randomly divided into 2 groups by dopamine agonists(QNP) modeling method to establish the rat model of OCD, a normal control group and QNP group. After behavioral testing, using ELISA method for the determination of serum BDNF and GDNF, using fluorescence method for determination of dopamine. Results 1. Patients with refractory obsessive-compulsive disorder have obvious cognitive impairment of language function than normal control group. 2. The level of serum BDNF and GDNF in patients with refractory obsessive-compulsive disorder was significantly lower than that of normal(P<0.01). 3. After modeling, the total distance of movement of the model rats was significantly increased, and the moving frequency was also significantly increased.Visit the sites and the duration of the not moving is significantly reduce, rotation was also significantly decrease,the duration of the central area were decreased, the central area frequency decreases. 4. The concentration of BDNF and GDNF in the blood of obsessive-compulsive disorder model rats was lower than that in the normal control group. 5. The concentration of dopamine in the cortex tissue of rats with obsessive-compulsive disorder was significantly decreased than that of normal rats,and the concentration of dopamine in the hippocampus no significant changes. ConclusionsPatients with refractory obsessive compulsive disorder have obvious cognitive impairment of language function, the abnormal concentration of BDNF and GDNF in the refractory obsessive compulsive disorder patients have no related with cognitive function, the concentration of BDNF、GDNF and dopamine in the rat of obsessive compulsive disorder have changed,may have a certain relevance with obsessive-compulsive disorder.
