Objective:The analgesic effect of epidural ropivacaine and fluorocitrate and related mechanismare investigated preliminarily in neuropathic pain rats model with chronic constrictiveinjury (CCI).Methods:72rats were randomly assigned to four groups as follows: group C, physiologicalsaline Infusion; group L, ropivacaine Infusion; group F, fluorocitrate Infusion; and groupLF, ropivacaine combine fluorocitrate Infusion. An epidural catheter was inserted into theepidural space at D7, and physiological saline,1%ropivacaine50μL,1nmol/μLfluorocitrate50μL,1nmol/μL ropivacaine combine fluorocitrate was administered at D8and D11, respectively. The mechanical allodynia of rats was assessed using a50%pawwithdrawal threshold (PWT) at D0, D7, D9, D11, D12, D14. Glial fibrillary acidic protein(GFAP) expression in spinal cord were analyzed by immunohistochemistry at D0, D7, D14.We analyzed the correlation between rates of PWT and GFAP expression at D0, D7, D14.Result:PWT, The PWT of each groups at D7reduced compared with that at D0. The PWT ofgroup C at D9, D11, D12, D14had no significance compared with that at D7. The PWT ofgroup L, group F, group LF increased, while group LF increased evidently compared withgroup L, group F, group L increased evidently compared with group F; Expression ofGFAP, the GFAP of each groups at D7increased evidently compared with that at D0, theGFAP of group C at D14had no significance compared with that at D7, while the GFAP ofgroup L, group F, group LF reduced, group LF reduced evidently compared with group Land group F, group L reduced evidently compared with group F; correlation analysis, the experimental results indicated that the negative correlation between the PWT and theGFAP, the PWT reduced with the increasing of the GFAP.Conclusion:Epidural infusion of ropivacaine, fluorocitrate or ropivacaine combine fluorocitratereduces neuropathic pain in rats model with CCI. Ropivacaine combine fluorocitrate showsthe best treatment effectiveness, ropivacaine shows the better effectiveness thanfluorocitrate. Ropivacaine reduces the neuropathic pain resulting from possiblysuppressing of glial cells activation in spinal cord.
