Objective:To discuss the effect and mechanism of c-fos expression in spinal dorsal neuron in neuropathic pain rat applying ropivacaine combined with sufentanil in sacral cannel injection.Methods:Forty chosen male Wistar rats were divided randomly into5groups: sham-operation group(J group),model comparison group (M group),ropivacaine group(L group),sufentanil group(S group),ropivacaine combined with sufentanil group(LS group).All rats were underwent the right sciatic nerve ligation expect the J group. The drugs were given via sacral cannel injection in post-operation from the1-14day. Recorded the pain behavior, mechanical hyperalgesia,thermal hyperalgesia. Detected the count of c-fos like immunoreactivity (FLI) cells using immunohistoche-mical methods.Results:(1) All rats were varying degrees ligated with valgus deformity and hind paw gesture of ground except the sham-operation group rats. The operation legs motions were in a good trend in L group、S group and LS group rats as time went by. A P<0.01was considered as statistically significant between LS group and M group about pain behavior in day14,No significant difference could be detected between M group and J group (P>0.05).(2) There were no significant differences among M group、L group、S group、LS group、J group about thermal hyperalgesia on no-operation legs (P>0.05). pre-operation and post-operation.All rats have behavior changes like paw withdrawal and hotplate.4operative groups reduced significantly compared with J group about thermal hyperalgesia and mechanical hyperalgesia on operation legs respectively and showed statistically significant (P<0.01).L group、S group、LS group decreased significantly compared with M group in day5、7、10、14respectively and detected statistically significant (P<0.05,P<0.01).The thermal hyperalgesia and mechanical hyperalgesia in L group、S group reduced significantly compared with LS group respectively (P<0.05,P<0.01) in day5、7、10、14.(3) The FLI cells of four CCI groups were mainly in Ⅰ-Ⅱ and Ⅴ-Ⅵ layer, and then Ⅲ-Ⅳ and Ⅶ-Ⅹ. M group、L group、S group decreased significantly compared with J group about c-fos expression in each layer (P<0.01).L group、S group、LS group decreased significantly compared with M group (P<0.01) on post-operation.The FLI cells of LS group reduced significantly compared with L group、S group (P<0.01)Conclusion:The sacral cannel applying ropivacaine combined with sufentanil had significant analgesic effect in neuropathic pain rats, and reduced the c-fos expression of the spinal cord cell.
