Objective:To explore the effects of mast cell degranulation on Serum Levels ofhs-CRP,ox-LDL and MMP-3in Atherosclerotic Mice and the Expression of MMP-3InPlaque.Methods:40Wistar male rats were radomly divided into four groups,which werecontrol group(group A),fed with normal diet,high-fat group(group B),Ket group(groupC) and OVA group(group D).The last three groups were treated with high fat diet andintraperitoneally injected of vitamin D3for establishing atherosclerotic animal models.Group C and group D were injected Ket and OVA respectively. Twelve weeks later,allof the animals were killed. blood was sampled from the hearts used to detectedhs-CRP、ox-LDL、MMP-3with ELISA method. The plaque pathologic morphology wasobserved with HE staining,and the the expression of tryptases and MMP-3in plaqueswere assayed with Immunohistochemical staining.Results:1.HE staining shows that the pathological changes of aortic atherosclerosis inthe group D were more severer than the group B and C, with intimal hyperplasia,foam cells accumulation and inflammatory cells infiltration.2.The level of serumhs-CRP in Group D was significantly higher than other groups (p<0.01). Group C waslower than group B (p <0.05);The level of serum ox-LDL in Group D was significantlyhigher than other groups (p<0.01). Group C was lower than group B (p <0.05);Thelevel of serum MMP-3in Group D was significantly higher than other groups (p<0.01).Group C was lower than group B (p <0.05).3.The distribution of mast cells anddegranulation of MC number were significantly increased in group D than other groups (p<0.01).Conclusions: Mast cell degranulation may have relationship with the plaqueunstabilization. |
