| The Tibetan medicine is being faced with lacking of germ-plasm resources andno perfect quality control system under the rapid development of the tibetan medicineindustry. In this paper, the development of SSR markers for Dactylorhiza hatagireaand study on quality control of Myricaria bracteata in Tibetan medicine were intro-duced in the first two parts. The last part summarized the innovation points and mainconclusion of the whole paper.There were two chapters in the first part, which mainly showed the developmentof mircosatellite(SSR) markers for Dactylorhiza hatagirea.The first chapter introduced the research status of the herbal medicine, applica-tion of DNA molecular markers and strategy in development of polymorphic SSRmarkers.The second chapter described the results of the method using magnetic beads toenrich the SSR markers from Dactylorhiza hatagirea.14polymorphic markers wereisolated and further characterized separately using68individuals from three widepopulations. The results showed the polymorphism information content (PIC)ranging from0.043to0.958and the average PIC were greater than0.5, indicating thepolymorph of the markers is rich. The14polymorphic microsatellite markers forDactylorhiza hatagirea provide powerful means to study the population geneticcharacteristics, protect the plant resources and evaluate the quality and so on.The second part divided into three chapters about the study on quality control ofMyricaria bracteata in Tibetan medicine.Firstly, an overview of Myricaria bracteata were introduced, including the plantgeographic distribution, chemical constituents, pharmacological activity and qualitycontrol. The research progress of flavonoids were also summarized in the thirdchapter. In the forth chapter, the content of quercitrin, quercetin, kaempferol andisorhamnetin were determined in12batches of the Tibetan medicine Myricariabracteata from different areas by phase high performance liquid chromatography(RP-HPLC) for the first time. Meanwhile, the content of four flavonoids in thedifferent organs were compared too. The method is rapid, simple and reliable.So it canbe used as an index in quality control of Myricaria bracteata.The cluster analysis of space geography and HPLC feature chromatography weredone for Myricaria bracteata of different parts on the qinghai-tibet plateau in chapter5. The results showed that the chemical composition has a difference even in the sameplace for medicinal material.Finally, the main conclusions of this two kinds of medicine study and theinnovation points were summarized in the third part. In the paper, the study aims toreflect the species resources and inner quality of the Tibetan medicine objectively andprovide the reference basis for the further development of Tibetan medicine andmedicinal materials quality control. |
