Xanthine oxidase catalyzes xanthine and hypoxanthine into uric acid.High concentration of uric acid can cause hyperuricemia, which may causegout. Inhibiting Xanthine oxidase activity, can prevent or delay the gout. Goutis a group of tissue injury diseases caused by uric acid increased because ofpurine metabolism, is the second largest metabolic diseases after diabetes.With economic development and improved living standards, the incidence ofgout increased year by year. In Chinese medicine, gout is similar to arthralgia.Eating out of control, alcohol abuse, and liking nutritious foods, which resultin loss of organ function, accumulate and cause gout. Western scholarsbelieve that the reason for gout is reduction of uric acid excretion caused bykidney disease or drugs, and (or) increased production of uric acid caused bygenetic defects of enzymes from nature and the environment or certaindiseases. So far, the food and drug can’t completely cure gout, only control it.Better clinical drug is xanthine oxidase inhibitor allopurinol, but there aremany side-effects. The Chinese medicines have broad sources, good treatmenteffect, few side-effects, which make it the anti-gout new drug research areasof focusing on.We selected traditional Chinese herbs for treatment of arthralgia andcontaining flavonoids, prepared a variety of extracts, Carried out inhibitoryactivity on xanthine oxidase in vitro, and determinated the kinetic parametersof five extracts which could have inhibitory activity. Then we made theethanol extracts of Semen Hoveniae that have the strongest inhibition intogranules, and carried out the peliminary Pharmaceutical Research of granules.Firstly, we study the preparation of the granules according to the physical andchemical properties of herb. Secondly, in order to control the quality effectively, content of total flavonoid in that ethanol extracts of SemenHoveniae granules were qualitative and quantitative analysised, and themethodology and the peliminary stability were also investigated, whichmake base for setting quality standards of the granules. And we carried out theinhibitory experiments of the granules on xanthine oxidase in vitro to confirmthe efficacy further.
