Purpose Using gene gun eukaryotic expression plasmid containing the BMP-2genelocal gene transfer in therapy of fresh bone defect. Methods Choose24healthy threemonths New Zealand white rabbits, respectively, set up new model of rabbit bone defect inmiddle of radial, grouping by the defect length:(1)8rabbits in1.5cm group,(2)8rabbitsin2.0cm group,(3)8rabbits in2.5cm group. Each group according to the interventionincluding:4rabbits in treatment group (BMP-2gene transfection group) and4rabbits inthe control group. Using gene gun for BMP-2local transfection after bone cuttingimmediately, the control group with natural healing after bone cutting. X-ray examinationsare taken In1week,3weeks,4weeks and9weeks after transfection, and the tissuesbetween or adjacent of the bone defect are taken to doing BMP-2Western blot detection,and radius specimens are taken for gross observation in1week,4weeks and9weeks,evaluation of the healing. Results Gross observation, X-ray examination and molecularbiology detection has a better consistency: treatment group is better than the control groupin bone healing(P<0.05). Conclusion Application gene gun mediated BMP-2gene to dolocal gene transfer in therapy of fresh bone defect is efficacious, and may be a effectiveapproach to cure delayed healing of bone and bone nonunion in the future.
