Objective: To explore the inhibitory effects of recombinant human endostatin (Endostar, ENDOSTAR) onretinal neovascularization of oxygen-induced retinopathy model in miceMethods: Sixty C57BL/6J mice were randomly divided into normal control group (A), high oxygen controlgroup (B group), saline control group (C), Endostar intervention group (D group), n=15, except thegroup A, all group were established the OIR model. Group A, group B were not treated, group C in the12d were given intraperitoneal injection of0.1mlNS, group D in the12d injection were givenintraperitoneal injection of rh.ES the0.1ml, after5days, the endotheliocyte nuclei of new vessels extendingfrom retina to vitreous in the tissue-slice by HE, vascular endothelial growth factor(VEGF) expression wereexamined by immunohistochemistry.Results: the endotheliocyte nuclei of new vessels extending from retina to vitreous of group A is0.97±0.04, group B is24.30±1.73, group B is22.58±1.54, gropp D is3.38±027.the endotheliocyte nucleiof new vessels extending from retina to vitreous of group B and C were much higher than that of group A, the differences were statistically significant (P <0.01); group D compared with group B and group C, therewere significant differences among them(P <0.01); group D compared with group A, there weredifferences among them(P <0.05);By immunohistochemical methods, group A: the retinal layers arrangedin neat rows, see weakly positive expression of VEGF, mainly located in the nerve fiber layer and innernuclear layer; hyperoxic group B: the retinal tissue is unnormal, a large number of VEGF is expressed, theinner membrane, nerve fiber layer and neovascularization endothelial cells express the most obvious. groupC: The retina tissue is disorderal, the result of the inner membrane, nerve fiber layer and neovascularizationendothelial cells is positive, a large number of VEGF is expressed; group D: VEGF expression is notobvious. Group B and group C, compared with group A, there was significant difference(P <0.01);group B and C compared with group D, there was significant difference (P <0.01). Group D compared withgroup A, there was difference (P <0.05)Conclusion: Retinal neovascularization in a mouse model can be inhibited by intravitreal injection of0.1mlEndostar which reveals that Endostar may have potential theraputic benefits in retinal vascular diseases.
