Objective:To observe whether there was a retinal neovascularization inhibition by systemic Avastin therapy,how the inhibitory effect and the change of kidney.Methods:Sixty 7-day-old SD rats were devided into six groups that include air control group,high oxygen control group high oxygen NS group,large-dosage group,medium-dosage group and small-dosage group.Fifty rats were putted into the environment with 80%oxygen for 5 days to establish models of retinal neovascularization.In the treatment group,SD rats were given in different concentration of 15.625mg/kg,31.25mg/kg,62.5mg/kg in the tail vein 0.1ml; we gave NS 0.1ml in high oxygen NS group.While in air control group and high oxygen control group,SD rats were not given any treatment.To observe the inhibition of angiogenesis was observed by the retinal fiat and retinal tissue-HE staining and kidney side effects were recorded by renal pathology HE staining.Results:From retinal fiat,There were large retinal neovascularization in the high oxygen contral group compared with in the air contral group,and the density and shape of retinal neovascularization were disordered.This result indicated that the animal model of Oxygen-Induced retinopathy with vascular proliferation was successful.Comparing with high oxygen NS group,regular distributions and reduced density of retinal neovascularization were observed in the Avastin treatment group.The number of the endotheliocyte nuclei of new vessels extending from retina to vitreous was less in the Avastin treatment group than in high oxygen NS group(P<0.001).Avastin treatment also compared among the three groups were statistically significant(except in small and medium doses groups,P<0.05,the rest of the group,P<0.01). Renal pathology show that renal tubular epithelial cell degeneration and necrosis,the basement membrane fracture;Renal tubular epithelial cell degeneration and necrosis,interstitial infiltration of inflammatory cells in medium and small-dosage group;The number of glomerular cells were increased and renal tubular epithelial cell were degenerated in large-dosage group.Conclusion:Systemic Avastin therapy could inhibit retinal neovascularization in rat in a dose-dependent manner,and with the increase in dose inhibition become more obvious.In medium and small-dosage group,the injury of kidney was small,the injury of kidney of large-dosage group was slightly serious.
