Pharmacodynamics Study Of Compound Xuelian Capsule

This research is to pharmacodynamic re-evaluate anti-inflammatory and analgesic effects ofCompound Xuelian Capsule and establish cattle Collagen-induced arthritis（CIA），to discuss further theaction mode of Compound Xuelian Capsule cure rheumatoid arthritis, to provide experimental basis forclincical application.Objective:（1） To re-evaluate anti-inflammatory and analgesic effects of Compound Xuelian Capsule.（2）Observe therapeutic action of collagen-induced arthritis rats，and investigate the mechanisms of therapeuticeffect by survey serum factor: TNF-α and PGE₂of CIA rats.Method:1.Anti-inflammatory action test: The anti-inflammatory effects were investigated by ear edema ofmice induced by xylene, the permeability of celiac blood capillary and carrageen induced white blood cells（WBC） movement in rat chest;2. The Acetic acid writhing was used to observe analgesic effect;3.adjuvant-induced arthritis in rats;4. The therapeutic action of Collagen-induced arthritis rats: Thetherapeutic effect was investigated by measure and observe the effect of Compound Xuelian Capsule on theankle swelling of CIA rats. Take the synovial tissue, make biopsy and observe the synovial histopathologyof rats;5. The inflation of the serum factor: The phlogistic cell factor TNF-α, PGE₂was detected byenzyme linked immunosorbent assay （ELISA） to investigate some of the mechanisms.Result:（1） Compound Xuelian Capsule could suppress mice ear swelling by xylene.（2） it could inhibitmarkedly the increase in abdomen capillary permeability in mice induced by acetic acid, lower thepermeability of celiac blood capillary.（3） It could markedly reduce the number of WBC in the thoraxextravasate of rats induced by carrageenan.2. Compound Xuelian Capsule could decrease the number ofwrithes of mice caused by acetic acid.3. Compound Xuelian Capsule could significantly inhibit theprimary and secondary footpad swelling of rats induced by the complete Freund’s adjuvant.4. thetherapeutic effect of Compound Xuelian Capsule on type Ⅱ Collagen-induced Arthritis Rats:（1） it couldsignificantly decreased the arthritis swelling of CIA rats in the groups of Compound Xuelian Capsule, startto give medicine on the14th day of establish the CIA model, the6th day of administration.（2） It couldimprove the synovial tissue pathological damage, compared with the CIA model group rat, it couldsignificantly reduced inflammatory cell infiltration and hyperemia of synovial tissue.（3） the level of TNF-αand PGE₂analyzed by ELISA was significantly lower in the medium and high dose group of CompoundXuelian Capsule than that of the model group （P<0.05，P<0.01）.Conclusion: Compound Xuelian Capsule has obviously effect of anti-inflammation and analgesia.Meanwhile compound Xuelian Capsule could inhibit the collagen-induced arthritis in rats, the mechanismaction probably related to inhibit the expression of the inflammatory mediators TNF-α and PGE₂.
